Objective:To investigate the pathogenesis of deep vein thrombosis(DVT) from the Nuclear factor kappaB /Inhibitory kappaB (NF-κB/IκB) and interventional mechanism of Chinese Herbs on DVT, in order to provide new idea for prevention and cure of DVT. Methods:In the experiment, uses the vena cava inferior ligation law to establish the deep phlebothrombosis big mouse model.Wistar rats were divided into six groups which included Xiao Shan Tong Mai high dose group, Xiao Shan Tong Mai low dose group, Haperin group, Xue Fu Zhu Yu capsule group, Sodium Chloride group, sham group. Then to observe the level of NF-κBP65 in Polymorphonuclear neutrophil(PMN) through immunocytochemical method, observe the level of NF-κBP65,IκBαin vascular endothelial cell(VEC), the level of Intercellular adhesion molecule-1(ICAM-1) in vein wall through immunohistochemical method and in situ hybridization method, observe the pathological character under the light microscope and the electron microscope. Results:Experiment shows that higher level of NF-κBP65 in PMNs and VECs of model group, lower level of IκBαin VEC of model group, higher level of ICAM-1 and PMN in vein wall and more severe pathological changes in model group.Xiao Shuan Tong Mai can effectively reduce the expressions of NF-κBP65,ICAM-1 and PMNs, improve the expressions of IκBαand the pathological changes. Conclutions: The system of NF-κB/IκB may participate in genesis and development of DVT, introduce the reciprocity of PMNs and VECs and injury of vein. The chinese herbs can intervene the system of NF-κB/IκB to suppress inflammatory reaction and thrombosis.
