Objective To explore the changes and significance of inflammatory cells and mediators in the process of acute traumatic venous thromboembolism by comparing the differences of the inflammatory cells infiltration in venous walls, plasma protein levels of CRP, TNF-α, IL-1β, IL-6 and IL-10, and verifying the relevance between the thrombus quality and other indicators in each group.Mathods 1, Established SD rat models of venous thromboembolism by the means of ligation and clamp of femoral veins, and then divided the model animals into 8 groups including group A, group B, group C, group D, group E, group F, group G and group H based on the differences of observing time and the conditions of thombosis in femoral veins; 2, Obtained the femoral veins segments of model animals for the weight and length measurements so as to calculate the thrombosus weight, then made them into parafin sections that were HE stained and histological observed in order to observe inflammatory cells infiltration in venous walls; 3, Collected blood plasma by the centrifugation of the model animals' blood and detected the protein levels of CRP, TNF-α, IL-1β, IL-6 and IL-10 in each groups by ELISA; 4, Compared the differences of the results between the other model groups and the control group(group A), and the relevance between the quality of venous thrombus and other indicators in each group; 5, Counted the results of group C, group E and group G as the group with thrombosus, group D, group F and group H as the group without thrombosus, and compared the differences between the two groups.Results 1, We got enogh 10 cases for each group during the observing time; 2, Compared with group A, there were significant changes of thrombosus quality in group C, group E and group G; 3, Compared with group A, there were significant changes of inflammatory cells infiltration in group B, group C, group D, group E, group F and group G, and there was no relevance between the differences of inflammatory cells infiltration and the thrombosus quality in each group; 4, Compared with group A, there were significant changes of the protein levels of CRP in all other groups, and there was no relevance between the differences of the protein levels of CRP and the thrombosus quality in each group; 5, Compared with group A, there were significant changes of the protein levels of TNF-αin group C, and there was no relevance between the differences of the protein levels of TNF-αand the thrombosus quality in each group; 6, Compared with group A, there were significant changes of the protein levels of IL-1βin group B, group C, group E, group F and group G, and there was no relevance between the differences of the protein levels of IL-1βand the thrombosus quality in each group; 7, Compared with group A, there were significant changes of the protein levels of IL-6 in group D, and there was no relevance between the differences of the protein levels of IL-6 and the thrombosus quality in each group; 8, Compared with group A, there were significant changes of the protein levels of IL-10 in group C, group E, group G and group H, and there was no relevance between the differences of the protein levels of IL-10 and the thrombosus quality in each group; 9, The differences of the results of thrombosus quality and protein levels of IL-1βbetween groups with and without thrombosus were statistically significant, distinguished from the other indicators including inflammatory cells infiltration in venous walls, the protein levels of CRP, TNF-α, IL-6 and IL-10.Conclusions 1, It is effectively to establish rat models of venous thromboembolism by the means of ligation and clamp of the femoral veins; 2, The difference of thrombosus quality between groups with and without thrombosus was statistically significant, so the caculation method of thrombosus quality by venous weight(or the thrombosus including)/venous length was realistic and useful; 3, The levels of inflammatory cells infiltration in venous walls, the protein levels of CRP, TNF-α, IL-1β, IL-6 and IL-10 were changing during the process of thrombosis, so they may be concerned with the process; 4, There was no relevance between the differneces of all indicators and of the thrombosus quality of each group, but the statistically significant differences of the results of thrombosus quality and protein levels of IL-1βbetween groups with and without thrombosus showed that IL-1βwas the most closely inflammatory mediator which was concerned with the process of thrombosis in all indicators; 5, The specific mechanism of the inflammatory cells and mediators involved in venous thromboembolism is unknown, the former may be the cause, influencing factors, or just the consequences of venous thromboembolism.
