Objctive: Deep vein thrombosis(DVT)is a important complications in patient with trauma or post-operation. To diagnose DVT is necessary since fatal pulmonary thromboembolism(PTE) may take place during the first 2 or 3 weeks after DVT occurs.Diagnosis of DVT may be difficult in clinic as patients with typical symptoms is less than 50 percentage. Activated platelets play an important role in venous thrombosis. Alpha-granule membrane protein-140(GMP-140 , or P-selectin) is a activation-dependent platelet receptor, which can reflect the level of platelet activation.Von Willebrand Facto(rvWF)is a sensitive marker of endothelial damage. GMP-140 and vWF can reflect the vein thrombosis in coordination. However, trauma or operation by itself can result in endothelial damage and inflammation, which may increase the levels of vWF and GMP-140 in blood. Interleuken-10 (IL-10) is a repressive cytokine, and the decreased IL-10 concentration in blood in the patients of trauma or operation may imply strengthen of the inflammatory reaction and increased probability of thrombogenesis. This study is to evaluate the vWF, GMP-140 and IL-10 in the diagnosis of DVT in patients with trauma or post-operation.Methods: The venous blood samples taken from DVT group(15 cases with trauma or post-operation), trauma group(18 cases with trauma or post-operation,without DVT) and control group(13 healthy cases) were detected for vWF, GMP-140 and IL-10 by enzyme linked immunoadsorbent assay ( ELISA) .Results:1 The blood levels of vWF were (49.031±29.730)%, (45.800±27.060)% and (22.900±9.600)% respectively in DVT group, trauma group and control group. The blood concentrations of vWF in both DVT group and trauma group were higher than that of control group( P<0.05), but no significant difference was found between DVT group and trauma group.2 The blood concentrations of GMP-140 were 64.021±17.594μg/L, 55.962±24.184μg/L,and 7.766±1.689μg/L respectively in DVT group,trauma group and control group. the blood levels of GMP-140 in both DVT group and trauma group were higher than that of control group ( P<0.01).3 The blood contents of IL-10 were 21.450±4.710 ng /L , 39.820±8.520 ng /L and less than 7.800 ng /L respectively in DVT group , trauma group and control group. Significant difference exsited for the IL-10 level not only between DVT group and control group ( P<0.01), trauma group and control group( P<0.01), but also between DVT group and trauma group( P<0.05).Conclusion:1 Trauma or operation can activate platelets and induce endothelial damage,which may result in increased blood levels of vWF and GMP-140.2 As a repressive cytokine during inflammation, IL-10 may be useful for evaluation of DVT formation in patients with trauma or post-operation.
