| The germination physiology, desiccation and storage tolerance of A.mongolicun Regel seeds with different chemical treatment, ultra-dry treatment and storage, processing and aging methods and rare earth element soaking were determined, and the relation physiological mechanisms would disguised, so the response physiological mechanisms of vigor and longevity of A.mongolicun Regel seeds in different treatments, the results would provide the reference to improving the vigor of seed and prolonging the longevity and providing gremplasm of A.mongolicun Regel.The results showed that:1. BTH had inhibition on the germination of A.mongolicun Regel. seed distinctly, but other soaking treatments had promotion on the germination, especially the soaking method with400mg·1Cecl3which could accelerate the germination speed and promote the quality of germination.Content of fat decreased at first and then increase with the increased of the germination days. The highest content of fatty acid were linoleic acid, and the oleic acid were secondly. The dynamic change of saturated and unsaturated fatty acid were adverse. The physiology and biochemical indexes changed little in35~40days of development.2. It would take43days the moisture content decreased from6.0%to1.5%. Suitable moisture content limit was3.5%and the content of MDA of the seeds with3.5%moisture content were lower than other moisture content, but the O2germination rate were higher than other moisture content at3.5%.3. The range of suitable moisture content would inlarge with the decrease of storage temperature. Meanwhile, all the germination indexes would reduce with the prolong of the storage time, especially the germination percentage of the seeds with6.0%moisture content stored for420days would decrease to2.5%, but the germination percentage of the seeds stored at35℃,20℃and-20℃would maintained at above85%. The content of MDA and O2-germination rate would increase with decreasing of moisture content. The activity of LOX, CAT, APX, PPO, GPX and GR stored at50℃were higher than other storage temperatures, and the activity of SOD, LOX, APX and GPX of seed stored at1.5%and2.5%were higher than other moisture.4. The germination indexes of A.mongolicun Regel seeds soaking with Ce with the moisture1.3%and2.3%stored at20℃,35℃and50℃conditions for eight months were higher than the same seed without Ce soaking. The activity of GR, LOX, APX, CAT, SOD and GPX of seed with Ce soaking were higher than the seed without Ce soaking, when the moisture were1.3%at20℃, and all the moisture at35℃, but only the activity of CAT of seed with Ce soaking were higher than the seed without Ce soaking at50℃.5. The effect of Ce3+soaking on germination indexes were non-significant, but Ce soaking have delayed effect on the declining vigor seeds for stored from3years to15years. The content of ASA and proline of soaking seeds were higher than without soaking seeds, but the content of soluble protein and heat-stable protein were lower. The activity of SOD, APX, GPX and LOX of seeds with Ce soaking were higher than without soaking, but the content of MDA and O2-germination rate were lower. It showed that Ce could increase antioxidant enzyme activity, thereby reducing harmful lipid peroxidation. |
