Studies On Ultra-dry Storage And Its Physiological-biochemical Changes Of Melon Seeds

In this research we select five conventional melon varieties seeds including Baishami,Hongchengcui,Balixiang,Huishuzi and Longtian as the material.They were tested by ways of ultra-dry storage.The changes of different vigor indexes of melon seeds were analyzed before and after storage,and suitable ultra-low moisture value and low moisture value of different melon seeds were found.1.The regression analysis of the melon seeds origination vigor targets indicate that melon seeds showed that the germination energy,germination percentage and germination index of seedling growth test and germination percentage of the cold test, were significantly correlated with simulated field condition,the correlation coefficient are 0.891,0.904,0.934 and 0.920 differently.The germination percentage of accelerated ageing is highly significantly correlated positively with seeding rate of simulated field condition,and the correlation coefficient is 0.991.According to the results of this experiment,the seedling growth test,accelerated ageing and cold test were the most proper methods for testing the seed vigor of melon.2.Melon seeds were dried by discolored silica gel as drier,and different moisture grades of five melon seeds were got.Two materials of each moisture grade were fetched,put,sealed and preserved for six months under normal temperature and low temperature(0~5℃) separately.Melon seeds before and after storage were tested by ways of seedling growth test, accelerated ageing test,cold test,electrical conductivity test,simulated field test, activation test of POD,CAT,SOD,TDH and APA and MDA content test.Compared the difference of different vigor indexes before and after storage,the results showed that suitable low moisture value and ultra-low moisture value of five melon seeds were found in normal temperature storage and low temperature storage,and there were differences among the varieties.Suitable low moisture and ultra-low moisture of Baishami under normal temperature storage were separately 6.07%and 4.09%,and suitable low moisture and ultra-low moisture of low temperature storage were separately 5.09%and 3.08%; Suitable low moisture and ultra-low moisture of Hongchengcui under normal temperature storage and low temperature storage were the same,and they were 6.09% and 3.03%separately;Suitable low moisture and ultra-low moisture of normal temperature storage of Balixiang were separately 6.03%and 4.09%,and suitable low moisture and ultra-low moisture of low temperature storage were separately 5.01%and 3.02%;Suitable low moisture and ultra-low moisture of normal temperature storage of Huishuzi were separately 6.08%and 3.03%,and suitable low moisture and ultra-low moisture of low temperature storage were separately 6.08%and 4.08%;Suitable low moisture and ultra-low moisture of Longtian under normal temperature storage were separately 6.03%and 3.06%,and suitable low moisture and ultra-low moisture of low temperature storage were separately 6.03%and 4.07%.After storing ten months with the storage of normal and cold temperature,several enzymes activation of melon seeds with each moisture,most moisture treatments are higher than the comparison or close with the comparison,the change rule of Baishami's ADH activation is not obvious,Hongchengcui's ADH activation all low compare with the comparison,the MDA content of every moisture grad in each varitety seeds is decreased highly.3.Fave melon variety seed in this research are suitable to carry on ultra-dry store, and can maintain the higher seed vitality...
