Aging is a kind of comprehensive phenomenon in the whole process of life,during which the organism’s morphology, structure and function will gradually decline and the skin aging is one of the indispensable important part. Skin aging is not only detrimental to people’s appearance, but also has a close pathological relationship with the occurrence of many skin diseases such as seborrheic keratosis,fluorescent keratosis, basal cell carcinoma, and squamous cell carcinoma.In the recent research studies, it is found that the effective components of Chinese herbal medicine can promote the synthesis of fibroblast collagen and elastin in human body,which can slow down the aging of cells. Among the Chinese herbal medicine,Ganoderma lucidum due to its functions such as immune regulation,scavenging free radicals, balance metabolism and so on, coupled with its mild property, becomes more and more popular. Ling Zhi-8(LZ-8) extracted from the mycelia of Ganoderma lucidum is a new immunomodulatory protein, whose structure is compact andcomponent single, but because of the difficulties in producing it, it has a low yield.Therefore, our research group chooses recombinant Ling Zhi-8(r LZ-8), to study r LZ-8’s effect on preventing skin aging.This research is to establish a model of aging by rats. And then the r LZ-8 was used to treat the aged rats by intraperitoneal injection, and then observe the water content, hydroxyproline(HYP), hyaluronic acid(HA), superoxide dismutase(SOD),malonicaldehyde(MDA), and the lipofuscin(LF) content of the skin tissue of each group of rats. After this, we can discuss the r LZ-8’s effects on preventing skin aging.Experimental results show that the water contentin the skin tissue of rats from the rlz-8 treatment group are significantly higher than those in the aging model group(P < 0.05), and the content of HYP and HA are also significantly higher than those ofthe aging model group(P < 0.05). It shows that rlz-8 not only can prevent water loss in rat skin but also can improve the hyaluronic acid content in the rats’ skin,promote the synthesis of collagen in the skin, thereby improving the elasticity of skin.At the same time,after the r LZ-8 treatment, the SOD content is much higher than those of the aging model group and the content of MDA and LF is much lower than those of the aging model group(P<0.05),which shows that r LZ-8 can enhance the superoxide dismutase SOD level in the body of the rats, reduce the lipid peroxidation products malondialdehyde content to achieve the purpose of antioxidation, decrease the level of lipofuscin in the rat, and then slow down skin aging. From the comparison of rlz-8 treatment group and model group, we can know that rats from the experiment group whose dermis become thick, collagen fibers and the gloss increase significantly, which illustrates that the rlz-8 can improve the degree of skin aging.In conclusion, r LZ-8 can effectively antagonize the aging of skin cells induced by D-galactose through increasing skin elasticity and improving the degree of oxidation.Data provided by this study will provide some scientific experimental basis for the research and application ofr LZ-8’s effects on preventing skin aging.
