Strengthening basic research on Chinese medicinal materials is the important way to improve the ability of original innovation in TCM industry. And it is the sufficient and necessary condition to achieve key breakthroughs in TCM research and development. Angelica dahurica radix is one of the40commonly used Chinese medicinal materials. Same as many other authentic medicinal materials, although there are many research reports about Angelica dahurica radix in recent years,but hardly any study on biological source mechanism of the active ingredients, especially relevant botany basic research. We previously showed that the coumarin was closely related to secretory tissues in Angelica dahurica.But up to now,there are few reports on the relationship.So the further scientific and systematic research is needed.The relationship between the coumarin and secretory tissues in Angelica dahurica will be systematically explained for the first time. This thesis will lay a strong base for the future research on biological source mechanism of the coumarin.The result will have an important role in revealing the key factors about medicine quality and yield of Angelica dahurica radix. At the same time, as the basic research, the results of this project has a positive meaning for the further studies on Angelica dahurica radix for achieving original innovation.Based on the previous studies, we studied the relationship between the coumarin and secretory tissues in Angelica dahurica radix at plant tissue and cellular level by introducing advanced technologies of structure botany and histochemistry, such as transmission electron microscopy(TEM), laser scanning confocal microscopy(LSCM). The main research work is as follows:1. Comparative anatomical study on structure and distribution of secretory tissues in different nutritive organs of Anglica dahuricaThe study results show that there are secretory tissues which the structure are similar in root,stem,leaf of Anglica dahurica. And the secretory tissues are most frequently located in the phloem of root which is the medical part of Anglica dahurica and located around the vascular bundles of stem and leaf. This paper mainly researches the anatomical characteristics of secretory tissues in root from distribution, structure and type. The results show that secretion cavitites are scattered in the inner bolt and secretion canals are higher abundance distributed with a racial, radial pattern in the phloem. 2. Study on ultrastructure of the secretory canals in the root of Anglica dahuricaA feasible preparation method of ultrathin slicing of Anglica dahurica root was finded out. The ultrastructure features of the secretory canals in the root of Anglica dahurica was observed and revealed by TEM.The electron microscope observations suggest that secretory cells in the Anglica dahurica root contain many plasids,endoplasmic reticulum, ribosomes, mitochondria and Golgi complexes,but much smaller plastids have lining structure. The quantity of mitochondrial crista are not affluence. In epithelial cells endoplasmic reticulum and ribosomes distribut in the the cytoplasm near the side of the seretory cavity.But the plastids in parenchyma have complicated lining structure and accumulate starch.3. Histochemical study of coumarin in the root of Anglica dahuricaIn this experiments a optimal cryo-section method in the root of Anglica dahurica is selected for the first time. The histochemical allocation of the coumarin in the root of the Anglica dahurica was studied by using the fluorescent microscopy. The results show that the coumarin, which releases blue fluorescence under the fluorescent microscopy, exists in secretary tissues. Meantime,relative content of coumarins in roots of Anglica dahurica were studied by using LSCM.4. Study on the relationship between the secretory canals index and the imperatorin contents in the root of Anglica dahuricaRelevance was analyzed between secretory canals index and imperatorin contents by using the method which is putted forward the measuring and calculating method of secretory canals index for the first time in this thesis. The result shows that there is a linear relationship between coumarins contents and secretory canals index on root in Anglica dahurica. Bigger the secretory canals index, higher the imperatorin contents.This thesis study Chinese medicinal material from biological source mech anism of the medical composition by using research methods of Chinese medicin e and botany, such as structure botany, histochemistry,medical botany, TCM identify,TCM chemistry.To study Chinese medicinal material by relating medical compositions and the structure of plant, to make the advantages of relevant departments complementary with each other, and to provide reference f or development of the TCM research.
