Objective:①To explore the protein expression of HIF-1α,MMP-9and CD34in normal brain tissues and various astrocytomas;②To analyz the expression of HIF-1α,MMP-9and CD34in astrocytomas;③To observe the correlation between HIF-1α,MMP-9,CD34and the malignant phenotype of astrocytoma.Methods:60cases of astrocytoma and9cases of normal brain tissues were stained with immunohistochemistry by PV-9000method on the same conditions to observe the positive expressions of HIF-1α, MMP-9and CD34. The normal brain tissues were acquired from decompression surgery in patients with intracranial hematoma, including hypertensive cerebral hemorrhage in4cases and aneurysmal hemorrhage in5cases. The60cases of astrocytoma were classified by WHO standard (grade Ⅰ in13cases, grade Ⅱ in15cases, grade Ⅲ in16cases and grade Ⅳ in16cases). The correlations between HIF-1α, MMP-9, CD34and the malignant phenotype of astrocytoma were analyzed by SPSS15.0.Results:1. Expression of HIF-la in normal brain tissues and astrocytomaExpression rates of HIF-1α in normal brain tissue and astrocytoma were respectively0.0%and73.33%, The differences was statistically signigicance between the two groups (P<0.01).Expression rates of HIF-1α in high grade astrocytoma and low grade astrocygoma were respectively96.88%and67.86%,the differences was statistically signigicant between this two guoups (P<0.01)2. Expression of MMP-9in normal brain tissues and astrocytoma Expression rates of MMP-9in normal brain tissues and astrocytoma were respectively0.0%and78.33%. The differences was statistically signigicant between the two (P<0.01). Expression rates of MMP-9in high grade astrocytoma and low grade astrocygoma were respectively87.50%and64.29%,the differences was statistically signigicant between the two groups (P<0.05)3. Expression of CD34in normal brain tissues and astrocytomaThe microvessel density were respectively40.54±5.20、50.41±3.82、60.25±4.55、77.45±6.32、38.24±2.21in different levels of astrocytoma respectively (grade Ⅰ, Ⅱ, Ⅲ and Ⅲ)and normal brain tissues. The differences among groups were statically signigicant (P<0.05)4. Correlation ananlysis between each factorThe expression intensity of HIF-1α and MMP-9, HIF-1α and MVD, MMP-9and MVD in astrocytoma were significantly positive correlated,the Pearson correlation coefficients were respectively0.841,0.624and0.701.Conclusions:1. HIF-1α and MMP-9were not expressed in normal brain tissues, but incomplete expressed in astrocytoma. The epression intensity increased with the malignancy of astrocytoma. Therefore, they may be involved in the invasion of astrocytoma and promote the invasion and growth of astrocytoma.2. The MVD increased with the astrocytoma malignancy, and was higher in astrocytoma than those in the normal brain tissues.bAccordingly, the relationship between MVD and astrocytoma was extremely close.3. HIF-1α and MVD, MMP-9and MVD were showing a positive correlation, indicating that HIF-1α and MMP-9were involved in astrocytoma neovasculatization.4. The expression intensity of HIF-1α and MMP-9in astrocytoma were positively correlated, suggesting that HIF-la and MMP-9may interacted with each other in the genesis of astrocytoma. They may lead the normal physiological state to disorder, thus helping to the proliferation and invasive of astrocytoma.They provided new targets for gene therapy of astrocytoma and a new criterion for diagnosing the grade of astrocytoma.
