ObjectiveBy detecting the different level of HIF - 1α,VEGF and MVD in 53 paraffin - embedded tumor tissue of human breast cancers by immunohistochemistry, we will find out where there are the significantly association among HIF - 1α, VEGF and MVD. We want to reveal the relationship between them and the different clinic characters and the relationship between them and prognosis. To find where HIF — 1α can be a marker of the prognosis in human breast cancer.Methodspecimens: a group of 53 paraffin — embedded tumor tissue of breast cancer patients at the first clinic college oncology department of China Medical University was used.All the tumor tissue were investigated by HE. the expression of HIF - 1α, vascular endothelial growth factor, Microvessel Density was investigated by immunohistochemistry. The results were drown out according to the original positive slices given by reagent corporation.Statistics analysis: SPSS 12.0 for window was used for analysis. The datas of MVD are presented as mean standard deviation (x ± s) , one - way ANOVA was used to compare the mean of each groups. T - test was used to compare ever-y two groups.DiscussionsAngiogenesis play a very important role in tissue repairing, inflammation and malignant tumors. During malignant tumors were growing up, local tissue must be anoxic for the speediness of tissue hyperplasia. By immunohistochemis-try.we can find HIF - lawas overexpressed in most malignant tumors cells,no expression was found in alkylic cells of tumor tissue and vicinal normal tissues. HIF - la was overexpressed obviously in the zones of necrotic tumor tissue.According to most studies, VEGF and bFGF were the two most important cell factors of angiogenesis. Transcriptional factor Hypoxia - inducible Factor -1 directly control the expression of VEGF in gene level, it was one of the primary control factors for malignant tumors to angiogenese.There were obviously relationship between the expressions of HIF - 1 a and tumor' s character and ability of invasion. By investigating of clinic tissue specimen we can find that the expressions of HIF -1 a were not high in normol tissue but they were distinctly high in tumors and precancerous conditions.ResultsNo association was found between HIF - la, VEGF and catamenia,pathology types ,ER ,PR( p <0. 05) in the whole group of the patients. The positivity of HIF -la, VEGF and MVD was strongly associated with tumor size, TNM stages, status of axilla nodes ( p < 0. 01). So there were positivity relationship between HIF - la and the prognosis.Conclusions1. The overexpression of HIF - la, VEGF and MVD were strongly associated with tumor size,TNM stages,status of axilla nodes.2. There were positive associations between HIF - la, VEGF and MVD.
