Research Of Different Syndromes In Senile Dementia Based On Metabonomics

Purpose： Searching the biological mechanisms of the syndrome of phlegmobstructing orifice,the syndrome of congealing cold with blood stasis,and the syndrome ofinsufficiency of kidney essence in senile dementia,then provides objective basis.for thetheory of the same disease with different syndromes in senile dementia.Methods:Analyzing the metabolite profiling diffferenee in blood and urine of ratswith three typical syndromes in senile dementia,which is the syndrome of phlegmobstructing orifice,the syndrome of congealing cold with blood stasis and the syndrome ofinsufficiency of kidney essence,searching the differences based on NMR in metabonomictechnology.Test biological indicators related to pathogenesis of this disease,combining withlocal hippocampal tissue pathological changes.Results:Compared with the normal group and the senile dementia,and compared witheach other,the blood and urine can be distinguished from each other in the three groupscombining with disease and syndrome,it certificates there are metabolite profilingdiffferenee between them.The results of hippocampal tissue pathology and biochemicalindicators showed pathological changes of the disease,there are no significant differencebetween the three syndromes groups except GST is significantly lower in the group withsyndrome of insufficiency of kidney,the group with syndrome of insufficiency of kidney isrelativly serious in terms of trends.Conclusion:Three are energy,lipid,glucose and amino metabolism disorder,andchange of organs functions,Tau protein hyperphosphorylation,neurotran-smitters reduced,peroxidation product increased in the three groups combining with disease and syndrome.These local balance destroyed eventually led to the "homeostasis" imbalance and causedisease.Relatively speaking,the disorder of bowel function and energy metabolism is serious in the group with syndrome of insufficiency of kidney essence,liver damage ismore serious in the group with syndrome of congealing cold with blood stasis.Combinedwith the overall metabolites,biochemical indicators and local tissue pathology results,itseems that the group with syndrome of insufficiency of kidney is more serious.It indicatesinsufficiency of kidney is the important factor in senile dementia.Different levels ofhomeostasis,and organ dysfunction different emphasis may be the same underlying causesof the same disease with different syndromes.
