Objective:To explore part of the micromechanism about the three typical syndromes of the senile dementia diseases, include the syndrome of cold coagulation and blood stasis, the syndrome of retention of phlegm and turbid substance in orifices and the syndrome of asthenia of kidney and deficiency of essence.Methods:Based on constructing the model of senile dementia using relevant theories of the modern medicine, combined with the theory of TCM etiology, using the method of combination of disease and syndrome, dynamic the stack factors, we constructing senile dementia animal model by the permanent bilateral common carotid artery ligation combined with long time continuous intraperitoneal injection of D-gal. Based on the model of disease, applying sexual overstrain and overfatigue, feeding high fat diet and cold stimulation, we established the senile dementia animal models combined the senile dementia disease with three typical syndromes. Then we observed behavioral and histopathology changes in animal models, to analysing of multiple biochemical markers related pathogenesis mechanism.Results:1. The senile dementia rats showed the declining of learning and memory abilities, changing include the hippocampus, the kidney and the testicular histopathological.2. Compared with the test results of the normalgroup, the test results of IL-6, TNF-α, MDA, GSH-Px, NO, SOD, Tau, Aβ1-42, AChE, ChAT, T and E2.3. Compared with the senile dementia group, in the syndrome of asthenia of kidney and deficiency of essence rat model, the active of GSH-Px, SOD, T, E2and ChAT decreased(P<0.05), NO increased(P<0.05) and the content of MDA increased(P>0.05); in the syndrome of retention of phlegm and turbid substance in orifices rats,the content of IL-6, TNF-α increased(P>0.05), Tau and Aβ1-42have shown a increase trendency (P>0.05); in the syndrome of cold coagulation and blood stasis rats, the content of NO increased(P>0.05), showing pathogenesis of multiple intersecting, mutual influence, a trend of collaborative pathogenicity. Conclusion:1. There are some pathogenesis of senile dementia:the activation of inflammatory cytokines, oxidative stress starting, modification and deposition of abnormal proteins, load of cholinergic system and decreas of sex hormone level. These pathogenesis can not only independent induct the changes in disease pathology factors, but also cooperative control the developing of disease.2. There are five kinds of pathogenesis cross-impact in the three typical syndromes of the senile dementia. The syndrome of asthenia of kidney and deficiency of essence related to oxidative stress starting, load of cholinergic system and decreas of sex hormone level. The syndrome of phlegm obstructing orifice related to the activation of inflammatory cytokines, modification and deposition of abnormal proteins. The syndrome of congealing cold with blood stasis related to the vascular injury and NO release.3. Combined with the overall metabolites, biochemical indicators and pathological results, seeing that the most serious pathological changes happen to the syndrome of asthenia of kidney, indicating that kidney diseaseis the basic pathogenesis of senile dementia is asthenia of kidney, the syndrome of asthenia of kidney and deficiency of essence is the typical syndrome of the senile dementia. |
