Objective: To study the biomechanics and effects of corresponding orincorresponding prescriptions to two typical AD syndromes Kidney Yang DeficiencySyndrome and Phlegm Blocking Orifices Syndrome in animals through behaviour study,pathological and biochemical findings.Methods:94rats were randomly divided into Control Group (ZC)10rats, AD ofKidney Yang Deficiency Syndrome (SA) group, AD of Kidney Yang Deficiency Syndrometreated by Jiawei Wendan Decoction (SW) group, AD of Kidney Yang DeficiencySyndrome treated by Shenqi Pill group (SS), AD of Phlegm Blocking Orifices Syndrome(TA) group, AD of Phlegm Blocking Orifices Syndrome treated by Jiawei WendanDecoction (TW) group, AD of Phlegm Blocking Orifices Syndrome treated by Shenqi Pill(TS) group with14rats in other groups. Two AD syndromes of Kidney Yang Deficiencyand Phlegm Blocking Orifices were induced in rats which were then gastrically fed witheither Shenqi or Jiawei Wendan Decoction accordingly. The biomechanism of ADsyndrome-prescription correspondence or non-correspondence were observed throughMorris Water Maze or optical microscopic section of hippocampus, kidney, adrenaline andliver or biochemical indicator of blood or hippocampus.Result:First, the result of water maze showed that, escape latency in the rest of the groupswas prolonged, resident time in target quadrant, the original platform across timessignificantly reduced except those in the ZC group and the difference was statisticallysignificant (P<0.05).Second, the findings of the optical microscope showed that the hippocampus, kidney, adrenaline and liver in Kidney Deficiency Syndrome groups of SA, SW and SS, thehippocampus and liver in the Phlegm Blocking Orifices Syndrome of TA, TW and TS weremore or less damaged. While the damages to hippocampus and other organs of the ratswere less severe in the prescription-syndrome corresponding groups than those in thenon-correspondence groups.Third, the blood biochemical indicators showed that TC, LDL-C increasedsignificantly in the Phlegm Blocking Orifices Syndrome groups of TA, TW, TS comparedwith the ZC group. TC LDL-C decreased significantly in the prescription-syndromecorrespondence group of TW. TC, LDL-C of rats in the prescription-syndromecorrespondence group of TS significantly increased (P<0.05) than those in TA group.T3, T4, CORT and T decreased in the Kidney Yang Deficiency Syndrome of SA groupand prescription-syndrome non-correspondence of SW group. No significant differencewas found in prescription-syndrome correspondence group of SS compared with ZC group.CD4+cell ratio increased and CD8+decreased in the Kidney Yang Deficiency Syndrome ofSA group compared with group ZC. CD4+cell ratio decreased and CD8+increasedsignificantly (P<0.05) in the prescription-syndrome correspondence SS group and theprescription-syndrome non-correspondence SW group compared with the SA group.Forth, biochemical indicators showed Tau protein, A β1-42, Bax and Bcl-2, AchE,MDA in hippocampus were increased significant in all groups other than ZC group(P<0.05). Those indicators in TA group were significantly lower than those in SA group(P<0.05). Those indicators in prescription-syndrome correspondence of SS and TW groupwere all lower than those in groups of the same syndrome.Conclusion:First, in this experiment, pathological damages in Kidney Yang Deficiency Syndromegroups were more severe than those in the Phlegm Blocking Orifices Syndrome groups.Pathological damages may present different severity in different syndromes of the samedisease.Second, prescriptions corresponding to the syndromes in this experiment can inhibithyperphosphorylation of Tau protein and abnormally increased Aβ1-42, and apoptosis inhippocampus, correct the cholinergic disorders of and oxidative stress abnormality in thisexperiment. It indicates that prescription-syndrome correspondence may have a multipletargets and multiple effects on disease.Third, prescriptions corresponding to the syndrome may correct pathological indexes of disease, such as the hippocampus related biochemical indexes. It also correct the relatedpathological factors of traditional Chinese medicine syndrome, namely the factors ofneuroendocrine system, immune system and blood lipid in this experiment. It implies thatprescription-syndrome correspondence can have a comprehensive and general effect.Forth, In this experiment, prescription of Shenqi Pill, which is not corresponding toPhlegm Blocking Orifices Syndrome, increased the pathological indexes TC, LDL-C.Jiawei Wendan Decoction, which is not corresponding to Kidney Yang DeficiencySyndrome partially correct the indicators. So prescription-syndrome correspondence mayhave two outcomes, one being totally invalid with all the indicators worsened and the otherbeing partially valid with certain target indicators improved and most left unchanged orworsened.
