| The liver is a vital organ that plays an important role in lipid metabolism,digestion,absorption,synthesis,decomposition,and transport.The mechanism of goose fatty liver formation induced by overfeeding with high-energy diets remains elusive;it is generally considered that fat synthesized in the liver,lipoprotein transport of fat,and destruction of the balance of fatty acidβ-oxidation are the main causes of abnormal deposition of fat in the liver,leading to the development of fatty liver.To establish the molecular mechanism responsible for fat deposition in goose liver,the cDNA sequence of the goose ApoA1,CD36 and DGAT2 genes were cloned and the pattern of gene expression which related to fat metabolism were studied by real-time PCR.The main results are as follows:1.The body weight,liver weight and their relationship,which have been breed100 days,and overfed 3 weeks,were analyzed in Landies geese.The results showed that the Landies goose live weight and body length,keel length,semi-submersible length were significantly associated(P<0.01).Liver weight and body length,keel length was significantly correlated(P<0.05).2.The cDNA sequence of the goose ApoAl,CD36 and DGAT2 were cloned by RT-PCR method and RACE,and the homology analysis showed that the cDNA sequences of the above three genes were highest homology with the duck,more than90%,higher homology with chicken,while with low homology with the human and other mammals.3.Ten 7-week-old healthy Landies ganders which with the same hatch time were randomly divided into two groups,overfeeding group and control group.The pattern of gene expression,including Spot 14,Apo A1,CD36,DGAT2,SREBP and OB,as well as the differences of gene expression between overfeeding geese group and the control group were investigated using real-time quantitative PCR.The results indicated that Spot 14 gene was expressed in all tissues,and with the highest expression in the abdominal fat tissue and intestinal tissue.The expression of Apo A1in adipose tissue,intestine and liver expression was significantly higher than in other tissues.The expression of CD36 gene in adipose and muscle tissues was higher than in other tissues.The highest expression of SREBP gene was in the kidney,the liver and the lungs have a certain amount of expression,but with a relative low expression in other tissues.The expression of DGAT2 gene in kidney,liver and lung tissues was higher than in other tissues.The expression of Spot 14 and Apo A1 genes in overfed goose liver tissues was significantly higher(P<0.05)than the control tissues,while significant differences were no found in other tissues.The expression of CD36 gene in the fat liver goose were significantly higher than the normal group(P<0.01),but no significantly different were found in other tissues.The expression of SREBP and DGAT2 genes in spleen tissue was significantly higher than the normal group(P<0.05),but no significantly different were found in other tissues. |
