Insomnia is the occurrence of sleeping or maintaining barriers, a kind of Syndrome of affecting the daily life, and belongs to disease areas of Mongolian medical Hei. Modern medicine to the treatment of symptomatic treatment, use more barbiturates, benzodiazepines and non benzodiazepines drugs to adjust insomnia. Drug treatment to improve the symptoms of insomnia, often accompanied by different degree of side effects, and easy to form drug dependence, curative effect and easy to rebound, is not an ideal treatment method. So it is important to find a safe, effective, no side effects, not easy to produce drug dependence treatment of insomnia. A large number of Mongolian medicine literature and modern clinical practice has proved that guided by the overall concept of Mongolian medicine warm acupuncture treatment insomnia is exact and reliable curative effect.ObjectiveThis study is that Mongolian medical warm acupuncture stimulate the PCPA induced insomnia in rats’Dinghui points、Hei points and Xin points as the starting point, and from the different levels, observe related cytokines before and after treatment of each group warm acupuncture、Monoamine neurotransmitters、amino acid neurotransmitters and other the variation of receptor-specific binding, clarify Mongolian medical warm acupuncture the hypnotic effect and mechanism for insomnia, and further verify the clinical practical value of the Mongolian medical warm acupuncture to treat insomnia.Methods60male SD rats were randomly divided into4groups, namely the normal group, model group, acupuncture group and the diazepam group,15rats in each group. Used the chronic intraperitoneal injection of amphetamine acid(PCPA) animal model of insomnia, Through the observation of Mongolian warm acupuncture indexes on the behavior of insomnia rats, brain DA, NE, Glu, GABA, Ach content and specific5-HT, the binding of DA to its receptor, to explore the Mongolian warm acupuncture treatment of insomnia rats and mechanism.Result1.Changes of body weight:compared with the model group, Changes of body weight of Mongolian Medicine Warm Acupuncture treatment group were significant differences(P<0.05);compared with other groups, body weight of the rat of the normal group. 2. Behavioral changes:the time of movement of rat of model group higher than normal group with highly significant differences(P<0.01);compared with model group, Mongolian Medicine Warm Acupuncture treatment group were significant differences (P<0.05).3.NE content:From the study on the influence on NE content of hypothalamus, hippocampus, pallium, showed that it can significantly reduce the NE content of hippocampus of rats of Mongolian Medicine Warm Acupuncture treatment group. Which were significantly different than that of model group(P<0.01); it can not significantly increase the NE of hypothalamus, pallium (P>0.05).DA content:From the study on the influence on DA content of hypothalamus, hippocampus, pallium, showed that it can significantly reduce the DA content of hypothalamus of rats of Mongolian Medicine Warm Acupuncture treatment group. Which were significantly different than that of model group(P<0.05); it can not significantly increase the DA of hippocampus, pallium (P>0.05).4. Glu, GABA^Ach content:From the study on the influence on Glu、GABA、Ach content of hippocampus, showed that it can significantly reduce the Glu content of hippocampus of rats of Mongolian Medicine Warm Acupuncture treatment group. Which were significantly different than that of model group(P<0.01); it can significantly increase the GABA、ACh content of hippocampus of rats of Mongolian Medicine Warm Acupuncture treatment group. Which were significantly different than that of model group(P<0.05).5.5-HT、DA Receptor binding rate:From the study on the influence on5-HT Receptor binding rate of hippocampus and pallium, showed that it can significantly increase the5-HT Receptor binding rate of hippocampus and pallium of rats of Mongolian Medicine Warm Acupuncture treatment group. Which were significantly different than that of model group(P<0.05); From the study on the influence on DA Receptor binding rate of hippocampus, showed that it can significantly increase the DA Receptor binding rate of hippocampus of rats of Mongolian Medicine Warm Acupuncture treatment group. Which were significantly different than that of model group(P<0.05); it can not significantly increase the DA Receptor binding rate of pallium (P>0.05).Conclusion1. Mongolian Medicine Warm Acupuncture can reduce the spontaneous activity, improve behavioral effects as reaction. 2. PCPA induced insomnia can lead to rat hypothalamus, hippocampus, prefrontal cortex monoamine neurotransmitter change, Mongolian Medicine Warm Acupuncture by reducing the NE content of hippocampus and hypothalamus DA content to improve sleep.3. PCPA induced insomnia can lead to hippocampal amino acid neurotransmitters change, Mongolian Medicine Warm Acupuncture could decrease the content of Glu in hippocampus, increase GABA contentand ACh content to improve sleep.4. Mongolian Medicine Warm Acupuncture on insomnia rat hippocampal5-HT, DA combined with the law and the prefrontal cortex of the brain5-HT binding law has obvious promotion effect.5. Mongolian Medicine Warm Acupuncture therapy to regulate the Heyi dysfunction, improve blood circulation, enhance immune function to improve the insomnia. And has the advantages of simple operation, safe, effective, less toxic side effect and other features and advantages.
