Based On IL17A/F-p38, The Mechanism Of Phlegm And Blood Stasis In Improving The Cerebral Blood Stasis Microenvironment Was Discussed

Objective:About one out of ten women suffers from chronic pelvic pain(CPP),which causes negative influences to people's normal life.Although modern medical science has developed three effective methods to treat CPP,such as symptomatic pain relief,hormone replacement,and surgical operation,these treatment methods cannot effect a radical cure for it.Therefore a search for the Traditional Chinese Medicine to cure CPP is quite necessary.The mentor put forward the theory that the blood stasis micro-environment results in CPP,based on which we analyze the mechanism of action of using Danzhi Decotion to remove blood stasis in order to cure CPP.Methods:In experiments,MTT method has been conducted to search for the best concentration of Danzhi Decotion and Aspirin to work on NIH/3T3 cells.By acting IL-17A/F on NIH/3T3 cells to obtain the concentration of the inflammatory factor MCP-1 and MCP-2,the best concentration of IL-17A/F was acquired where Danzhi Decotion has effect on P38 pathway and inflammatory factor MCP-1 and MCP-2.Therefore the mechanism how Danzhi Decotion treats CPP is proved.In clinical research,patients who suffer from Qi stagnation and blood stasis type of adenomyosis(CPP)are treated with Danzhi Decotion,compared with those treated with Sanjie analgesic capsule,to observe the function of Danzhi Decotion in effecting TCM syndrome score,CRS,CMSS.Results:In experiments,1.25%solution of Danzhi Decotion and 50ug/ml solution of Aspirin has less than 10%cytotoxicity to NIH/3T3 cells.The best concentration of IL-17A/F is 100ng/ml to obtain the concentration of the inflammatory factor MCP-1 and MCP-2,and both Danzhi Decotion and Aspirin inhibit the induction of IL-17A/F into NIH/3T3 cells to secrete MCP-1/MIP-2.Danzhi Decotion and Aspirin can inhibit IL-17A/F-P38 inflammatory signal pathway.Based on NIH/3T3 cells,IL-17A/F can stimulate P38 inflammatory signal pathway,while Danzhi Decotion and Aspirin inhibit IL-17A/F-P38 signal pathway.In clinical research,Danzhi Decotion is used as the experimental group,while the other TCM Sanjie analgesic capsule is used as the control group.The experimental group and the control group has statistic difference in the total efficiency related to TCM symptoms,and both the treatment and the total score of improving depression,anger and frequent sighing.In terms of NRS and CMSS,the experimental group has statistic difference from the control group in total marked effective rate,when the experimental group exhibits a better score than the control group in both NRS and CMSS.Both the experimental group and the control group have no statistic difference in uterine volume and CA125 before or after the treatment.No patients in either experimental group or the control group have adverse reactions.Conclusion:By inhibiting IL-17A/F-p38 and reducing the secrete of MCP-1 and MIP-2,Danzhi Decotion can alleviate CPP.Danzhi Decotion can effectively improve the TCM symptoms and clinical symptoms of patients who suffer from adenomyosis(CPP).
