Objectives:This study focuses on a biological process named epithelial qualitative transformation(EMT)occurring at the initial stage of tumor metastasis.We tested the change of migration ability after treating lung cancer lines with SPS(safflower polysaccharide).Then PCR Array assay whose gene list was acquired from Qiagen webside was performed to reveal the alteration of EMT-related genes.The objectives of this study is not only to determine the molecular mechanism of SPS but also widen the application of SPS,and provide a potential usage of SPS as an assistant anti-cancer drug through cellular and molecular exploration as well.Methods:1.Through Transwell chamber and cell scratch test(wound healing),observe the migration change multiple lung cancer cell line H460,H1299 and A549 after treating with safflower polysaccharide.2.Through PCR array technology,79 genesinvolved in EMT process was detected,the molecular map of EMT in SPS treating cells was identified.3.SiRNA sequence transtection experiment-towards E-Cadherin was performed to confirm whether the inhibition migration induced by SPS was due to the transcriptional changes.results:1.SPS can effectively inhibit the migration and mobility of lung cancer cell lines.2.PCR array experiment shows that the statistically significant changed genes which are all up-regulated in EMT in cells treating with SPS was decreased excep TMEM132A.Some genes down-regulated in EMT were showed to be increased at the same time.It showed that the epithelial characteristics of cells were enhanced,and the metastasis tendency was decreased obviously after the treatment of safflower polysaccharide.3.Several tumorigenesis-related genes involved in development and metastasis have been significantly changeds,such as E-cadherin,snail,EGFR and STAT3,The genes regulation by SPS showed a benign phenotyp.Besides,EGFR is one of the most classic targets for cancer therapy.4.After interference of E-Cadherin,migration inhibited cells treated by SPS showed a rescued migration ability which revealed that the change of migration was due to the transcriptional regulation.Conclusion:1.The study explores the effects of SPS on the migration and motility of lung cancer cells for the first time.2.According to PCR Array results,we found that the migration change caused by SPS comes from the regulation of EMT related genes such as E-Cadherin.3.Otherwise,SPS can also inhibit the mRNA expression of some oncogenes.These effects make SPS to become an effective antitumor drug.This study also gives an example on traditional Chinese medicine research.Provide a more in-depth understanding on SPS and its application on cancer treatment.
