Safflowers are the safflower's dried flower belong to copositae plants,planted in many places and mainly in XingJiang,HeNan, ZheJiang,SiChuan.The present study shows that the safflowers can be used as drug and food.They are considered as the ideal food additive for their natural edibile yellow pigments and red pigments and used as staining materials of cosmetic and textile fabric.They posses the effect of anti-cancer,disinfection,detoxication,depressurization and skin protection.There are many reports on yellow pigments at home and abroad but few at safflower polysaccharides.So the dissertation systematically studied the extraction technology,isolation and purification,composition analysis,molecular weight and initial pHarmacological action.It aims to find out a new active component and to develop the valuable resource of Safflower.The main contents and conclusions are as follows:1.To study the extraction conditions of polysaccharides by using water.The conditions of extraction were optimized by orthogonal experiment on the basis of singal factor tests.The result showed that the optimal conditions was as follows:the time is 1.5h,solid-liquid ratio is1:20,The extraction temperature is 80℃and the extraction times is 3 times.The effect order of the four factors for the extraction solid-liquid ratio,time,extraction times,temperature,and in turn.the content of polysaccharides was 25.57%.2.In order to study the purification technology of safflower polysaccharides.Three deproteinization method was compared.the optimal method was proved to be a combination of enzyme and Sevag. The conditions of decoloring method were optimized by orthogonal experiment on the basis of singal factor tests.The result showed that the optimal conditions was as follows:the dosage is 20%,pH is 8,the temperature is 60℃and the decoloring time is 180 minutes。Then they purified by DEAE-52 cellulose column chromatograpHy and Sephadex G-100 column chromatograpHy,received SP-1 and SP-2.3.study the purification technology of safflower polysaccharides. After the deproteinization,decolourisation,dialysis and DEAE-52 column chromatograpHy,we got two compositions(SP-1 and SP-2),the purify of them were 70.7%and 72.57%,respectively.HPLC and GC were used to determine the molecular and the compose of monosaccharides.The result were as follows:The molecular of the five polysaccharides compositions were 5.13×103 and 3.70×104.The monosaccharides compose of SP-2 were mannose,glucose, galactose;molar ratio was 8.4:3.7:1.the monosaccharides compose of SP-2 were glucose,mannose,xylose,rhamnose,molar tatio was 21.57:7.1:1.64:1.4.To observe the regulatory effect of Safflower polysaccharides on the growth of H22 tumor and the expression of VEGF and Ki67 in transplanted H22 tumor of mice by tumor weight,spleen index,thymus index and the expression of VEGF and Ki67 which was determined by the immunohistochemical method.It shows that the high dose group had significant effect on inhibiting the tumor growth and the other groups had degression trend.Safflower polysaccharides could make the expression level of VEGF and Ki67 down.
