Research backgrounds:Zanthoxylum nitidum(Roxb.)DC.is the dry root of Radix Zanthoxyli which comes from rutaceae Zanthoxylum bungeanum genus and it was recorded in the 2010 edition of "Chinese pharmacopoeia".Z.nitidum is the principal raw material of many famous Chinese patent medicines,such as Sanjiuweitai,Diedawanhua oil,Bonesetting liquid and the daily necessity of LMZ medicinal toothpaste and so on.Nowadays,the demand quantity of Z.nitidum has more than 4000 tons each year,and presenting a significant growth trend.The supplies of Z.nitidum are mainly through wild plants for years,root is the traditional medicinal part,because the regeneration ability of Z.nitidum is weak,it has encountered the bottleneck of resources problem nowadays,which seriously affected the proprietary Chinese medicine and its related industry raw material supply.The previous research of different medicinal parts of Z.nitidum's HPLC/MS/UV fingerprint showed that:the chemical composition of root and stem of Z.nitidum is similar;And use the HepG2/STAT3 activity test of high-throughput screening found the same chemical component of root and stem--8-me-thoxydihydrochelerythrine has a strong effect on the STAT3 which is the important member of Signal Transducer and Activator of Transcription Protein family,but its mechanism is unclear.In preliminary comparative study also found that the extracts from different parts(roots,stems,leaves,above ground)of Z.nitidum all have certain analgesia and gastric mucosal protection effects.Through literature survey,the other parts of Z.nitidumhave relevant records and medicine application.All of above suggests that the other parts of Z.nitidum especially the stem,have medicinal value and it is worthy a further development and utilization.The content of nitidine chloride was thought as a standard to evaluate the quality of Z.nitidumby the "Chinese pharmacopoeia".But a lot of research shows that the main therapeutic effects of nitidine chloride were anti-tumor,myocardial protection and so on.While Z.nitidum was used in a variety of inflammatory diseases on clinical.Inflammation is one of the main efficacies,nitidine chloride does not reflect the anti-inflammatory efficacy of Z.nitidum..So looking for the effective components which can directly related to anti-inflammatory with Z.nitidum is necessary.Objective:We compare both the chemical composition and medicinal efficacy of the root and stem of Z.nitidum,and study the mechanism action of the same chemical component from root and stem,Research the medicinal value of stem from the material foundation to efficacy validation to study the medicinal value of Z.nitidum,and exploring the mechanism of anti-inflammatory of the common chemical component.This can not only provide proof why ancestor use the other parts of Z.nitidum as a medicine,but also supply reference to expanding the medicinal part application and sustainable comprehensive exploitation and utilization of Z.nitidum.Otherwise lay foundation to make the stem of Z.nitidum as a medicine,finally realize the sustainable utilization of resources of Z nitidum.Protect the resource balance and ecological environment,meet the requirements of sustainable development of the traditional Chinese medicine industry,achieve good social and economic benefits,meet the needs of current economic and social development,and achieve a win-win situation finally.Methods:After making sure that the chemical composition of stem and root of Z.nitidum is similar.Then using liquid chromatograph/mass spectrometer,comparing the standards and active ingredients of medicinal material,which were preliminarily separated from the guide of biological activity in Z.nitidum.The same chemical components of root and stem were confirmed and their contents were measured.Then the quality compare was also studied between the root and stem of the Z.nitidum.In the other way,by observing the regulation of 8-me-thoxydihydrochelerythrine which has a strong effect on the STAT3 to the signal pathway of tyrosine kinase2/signal transduction and transcription activating factor 3.and the influence on the inhibit proliferation and induce apoptosis,cycle distribution of Gastric Adenocarcinoma cell(AGS)to clarify the anti-inflammatory efficacy material of Z.nitidum.According to the attending functions of Z.nitidum recorded in the pharmacopoeia,further pharmacodynamics comparison between root and stem were carried on from anti-inflammatory and analgesic,damage resistance,resistance to peptic ulcers,improvement the action such as gastrointestinal function and so on to study the medicinal value of the stem part.Results:We identified the same chemical components of root and stem were nitidinechloride,chelerythrine,8-me-thoxydihydrochelerythrine,oxychelerythrine,magnoflorine and neoherculin.We also investigated the morphological identification,moisture contents,ash contents and extract content contents of the root and stem.Compared with root,stem has longitudinal wrinkles and scattered stud or stud marks and pith.Both the moisture contents of root and stem are within 10%;The root' s ash contents are more than stem while the extract contents and the chemical composition contents were lower than with the roots of a batch.Efficacy results show that compared with the root of Z.nitidum,the stem also has strong pharmacological activities on anti-inflammatory and analgesic,anti injury,gastric mucosa protection and promote gastrointestinal motility,and the effects' strength of stem has little difference with the root.So the stem of Z.nitidum has a good medicinal value,which is worthy of further development and utlization.The same chemical component of root and stem--8-me-thoxydihydrocheler ythrine has a strong effect on the Janus kinase2-signal transductor and a ctivator of transcription3 pathway,It can inhibit JAK2 and STAT3 phospho rylation,also has a strong inhibit proliferation and induce apoptosis of AGS cells.Conclusion:Thus the chemical composition of stem and root of Z.nitidum are similar,only differ in their contents.The same chemical component of root and stem 8-me-thoxydihydrochelerythrine has a strong pharmacological activity on STAT3 which was abnormal expression in most malignant inflammation and tumor tissues.The function of 8-me-thoxydihydrochelerythrine may be by adjusting the activation of JAK2/STAT3 pathway related protein to realize the malignant inflammation and tumor cell proliferation inhibition.So 8-me-thoxydihydrochelerythrine may be the main anti-inflammatory efficacy of Z.nitidum.Through several efficacy of models can infer that stem of Z.nitidum has the similar pharmacological effects as root.And the strength difference is not obvious,those can ensure that the stem of Z.nitidum also have a strong medicinal value and can be considered as an alternative medicine to expand medicinal parts of Z.nitidum.Advocate stem of Z.nitidum who has a similar chemical composition,pharmacological effect,certain folk medicine amplification history as a medicine,increase medicinal part and its resources of Z.nitidum can relieve or resolve the resource scarcity problem.
