Objective: The lncRNAs were analyzed by the results of lncRNA microarray in Xinjiang Uygur and Han populations between Coronary Heart Disease and Health.The lncRNAs were screened from lncRNA microarray.The results showed that THP1 foam cell model and HUVEC endothelial injury model were established respectively.The expression of lncRNAs was observed by transfection of lncRNAs siRNA,and the mechanism of different lncRNAs in the development of coronary heart disease was discussed.Methods: According to the inclusion and exclusion criteria,lncRNA gene chip was used to detect the expression of lncRNAs in coronary heart disease and healthy control group,and the results were analyzed by gene chip analysis,signal pathway analysis and predictive results of target genes.After amplification of the sample size,the expression of lncRNAs in the anticoagulated peripheral blood of Uygur and Han were screened by real-time fluorescence quantitative PCR.Using two independent samples of the t test,statistical software SPSS 18.0 for statistical data processing.The expression of lncRNA was detected by real-time quantitative PCR.The expression of lncRNA was detected by transfection reagent.Real-time quantitative PCR detected the lncRNA expression level changes in two kinds of cells?HUVEC and THP1?before and after transfection of lncRNA siRNA.Results: The lncRNA expression profiles of Uygur and Han patients with stable angina pectoris and acute myocardial infarction were established by analyzing the results of lncRNA gene chip.A total of 1871 up-and 231 down-regulated lncRNAs were identified to be significantly and differentially expressed between Xinjiang Uygur SAP group and the healthy control group,and 795 up-and 554 down-regulated mRNAs were identified.Differences lncRNAs NR037652.1,ENST00000607654.1,ENST00000589524.1 and uc004 bhb.3 verify the results consistent with the chip test results.Compared with the healthy control group,there were 158 upand 384 down-regulated lncRNAs in Han SAP group,and 106 up-and 346 downregulated mRNAs between the two groups.Differences lncRNAs ENST00000418539.1,ENST00000589524.1,NR027469.1 and NR046226.1 were verified by qRT-PCR that the results were consistent with the chip test results.Compared with the Han healthy control group,there were 2677 up-and 458 down-regulated lncRNAs in Han AMI group,and 1168 up-and 1334 down-regulated mRNAs between the two groups.The expression of lncRNAs ENST00000509938.1,ENST00000581794.1,NR047662.2,uc002 ddj.1 and uc002 mev.3 were confirmed by qRT-PCR,and the results were consistent with those of microarray.The expression of different lncRNAs in cell levels was verified by qRT-PCR through establishing HUVEC endothelial injury model and THP1 foam cell model.Compared with THP1,lncRNA MAIT,MALAT1,uc002 ddj.1 and ENST00000509938.1 were up-regulated,and H19,ANRIL,NR-047662.2 and ENST00000581794.1 were down-regulated in foam cells.Compared with normal HUVEC,the expression of ANTIL,NR-047662.2 and ENST00000509938.1 were up-regulated after ox-LDL induction,and the differences were statistically significant?P < 0.05?,and the expression of ANTIL,MALAT1,H19,uc002 ddj.1 and ENST00000581794.1 were down regulated.The lncRNA siRNA was transfected into lncRNA siRNA,and the cells were transfected with lncRNA MAIT,MALAT1 uc002 ddj.1 and ENST00000509938.1 siRNA group compared with the control group.LncRNA MAIT,MALAT1,uc002 ddj.1 and ENST00000509938.1 The expression of siRNA was significantly decreased,the difference was statistically significant.Conclusion: The lncRNA expression profile of stable angina pectoris and acute myocardial infarction in Uygur and Han Chinese patients was established by analyzing the lncRNA gene chip.The lncRNA expression was consistent with the results of the microarray test,and could be used as diagnosis biomarkers of coronary heart disease.
