BackgroundProtein 4.1 has tumor inhibitory activity,through the FERM domain of tumor growth control firmly control,but the relationship between protein 4.1R and tumor is not clear,and lung cancer,especially the occurrence and development of lung adenocarcinoma is related,Remains to be resolved.PurposeTo investigate the relationship between the expression level of protein 4.1R and the prognosis and pathological features of lung adenocarcinoma.MethodsThe expression of protein 4.1R in lung adenocarcinoma was analyzed comprehensively by using two major databases: Oncomine and HPA.The expression of protein 4.1R was confirmed by commercial tissue microarray detection in 75 cases of paraffin tissue of lung adenocarcinoma,The main indicators of the analysis of protein expression-related factors,survival-related factors analysis.ResultsThe Oncomine website currently has 715 datasets,86733 samples.Statistical analysis of 31 lung cancers in 13 data sets involved protein 4.1R,and only one of the analyzes of protein 4.1R changed,compared with normal tissue,the overall expression of protein 4.1R in lung cancer was down-regulated.But the protein 4.1R expression in each data set is not exactly the same.The expression of protein 4.1R in lung adenocarcinoma is not yet conclusive,and the down-regulation is dominant.There are two data that support down-regulation of protein 4.1R expression.Bhattacharjee results showed that a totalof 203 patients had the most significant down-regulation of small cell lung cancer,4.523 times lower than normal lung,with a p-value of 7.23E-5,a significant difference in adenocarcinoma 1.798 times and a p-value of 0.021 The Weiss data showed that the expression of protein 4.1R in lung adenocarcinoma was 1.030 times lower than that in normal lung tissue,and the p value was 0.012,which was significantly different.Compared with normal lung tissue,the expression level of lung adenocarcinoma was 1.459 times and p value was 2.27E-9,which was significantly different from that of normal lung tissue.Hou increased 1.120 times,Selamat 1.091 times,Su 1.211 times,p value less than 0.05,there were significant differences.The data of up-regulation or down-regulation of protein 4.1R were:Okayama,Hou,Garber,Landi,Weiss,Beer,Bhattacharjee,Stearman,TCGA,p values were greater than 0.05,the results were not significantly different.From the above results,the expression level of protein 4.1R can be divided into two categories:up-regulation and down-regulation: protein 4.1R expression level is divided into two types: high expression and low expression,some samples are down,some samples are up,such as Weire Lung,Garber Lung,Selamat and other data sets.Big data analysis from HPAThe level of protein 4.1R gene expression in normal lungs was moderate.Protein4.1R HPA large data analysis results only gene expression results,there is no protein expression data.In general,the expression level of protein 4.1R gene was from high to low: the higher expression of bone marrow and immune system;the lower expression of tissue: brain tissue,pancreas.The level of protein 4.1R gene expression in the lungs is between the two.In the HPA database,the highest expression of protein 4.1R was lymph nodes,tonsils,bone marrow,appendix and spleen,and the content was higher than 20 FPKM.The tissues with low expression were: pancreas(2.4TPM),epididymis(4.3TPM),cerebral cortex,Skeletal muscle,myocardium,content less than 5 FPKM,other tissue expression between 5-20 FPKM,lung expression of 19 FPKM,are moderate levels.The expression level of the GTEx database is as follows: the highest tissue is the cerebellum,followed by the small intestine,the least(followed by low to high)is the hippocampus,the cerebral cortex,the myocardium.The expression level of lung was 3.2FPKM,and the expression level of other tissues was normal.In all chromosomes,protein 4.1R expression was present at both protein and expression > 10% was divided into high expression group.The total expression number of protein 4.1R was 72,including 1 of low expression,no death;and 71 peopleof high expression with 21 death.The P value was 0.0548,the difference was not significant for the survival time between two groups.The survival analysis for patients with lung adenocarcinoma based on the expression of protein 4.1R in the adjacent cytoplasm around tumor: In the 75 cases of paracancer cytoplasm,49 cases had lower expression of protein 4.1R with 19 cases of death,and 24 cases higher expression with 3 cases of death.P value was 0.041 and the difference was significant for the lower and higher expression groups.The results showed that the survival rate of patients based on protein 4.1R expression in the adjacent cytoplasm was higher than that in the tumor cytoplasm,indicating that protein 4.1R had tumor inhibitory effect,which was consistent with the literature.Why the adjacent tissue will have more protein correlation between the positive rate of expression,sex,age,tumor size,T stage,lymph node group and so on(P = 0.049-0.979).T staging and N shift 2,the correlation coefficients between TNM staging were 0.422 and 0.690,respectively,and the P value was 0.000,the correlation difference was significant.In addition to gender,tumor size,T stage and lymph node group,the pathological grade and cytoplasmic protein 4.1R expression positive rate,adjacent tothe cytoplasmic protein 4.1R expression positive rate,the nucleus of the nucleus There was no statistically significant difference in the correlation between the positive rate of protein 4.1R expression,age,N metastasis,and TNM staging(P =0.182-0.400).The correlation coefficients of the pathological grade and sex,tumor size,T stage and lymph node group were-0.37,0.365,0.269,0.336,and the P values??were 0.001,0.002,0.019 and 0.004,respectively.The correlation was significant.ConclusionsMeta data show that protein 4.1R is had the moderate level expression in normal lung tissue,with down-regulation mainly in lung adenocarcinoma tissue,but some of upregulation.Tissue chip detection showed that the sutvival time was longer for lung adenocarcinoma patients with higher expression of protein 4.1R in paracaner cytoplasm rather than cancer cytoplasm in 75 cases patients with lung adenocarcinoma and shorter survival time for lower expression.The expression of paracancer expression protein 4.1R rather than ttat within cancer itself was positively correlated with the survival time/ prognosis of patients with lung adenocarcinoma.
