ObjectiveTo detect the expressions of Caveolin-1,EGFR,β-catenin in lung cancer progression tissue microarray by immunohistochemistry and discuss their roles in lung cancer genesis and progress.With these results,we wanted to put our discoveries into the clinicopathological diagnosis,prognosis evaluation and gene therapy.Material and MethodWe used SP immunohistochemistry method to detect the expressions of Caveolin-1,EGFR andβ-catenin in tissue microarray with 270 cores,and analyzed the relationship between their expressions and clinicopathological parameter.Results1.The positive rate of Caveolin-1 in primary lung cancer and precancerous lesion were lower than that of normal lung tissue(P<0.05).The positive rate of Caveolin-1 was higher in the group ofⅢ+Ⅳstage thanⅠ+Ⅱstage(P<0.05), higher in the group with lymph node metastasis than without lymph node metastasis(P<0.05).2.The positive rate of EGFR in primary lung cancer was significantly higher than normal lung tissue(P<0.01).The positive rate of EGFR was significantly lower in SCLC than NSCLC.The positive rate of EGFR was higher in the group ofⅢ+Ⅳstage thanⅠ+Ⅱstage(P<0.05),it was higher in the group with lymph node metastasis than that without lymph node metastasis(P<0.05).3.The positive rate ofβ-catenin in primary lung cancer and precancerous lesion was lower than normal lung tissue(P<0.05).The expression ofβ-catenin was higher in the group of high-middle differentiation than low-un differentiation(P<0.01).The positive rate ofβ-catenin was lower in the group ofⅢ+Ⅳstage thanⅠ+Ⅱstage (P<0.05),and it was lower in the group with lymph node metastasis than that without lymph node metastasis(P<0.05).4.There was significantly negative correlation between EGFR and Caveolin-1,β-catenin(P<0.05,P<0.01 respectively).There was significantly positive correlation between Caveolin-1 andβ-catenin(P<0.01). Conclusions1.Caveolin-1 had dual effects on tumors.The positive rate of Caveolin-1 was lower in primary lung cancer and precancerous lesion than that in normal lung tissue, which proved that Caveolin-1 was a tumor inhibitor.The deletion of it can promote the genesis and progress of lung cancer.But the expression of Caveolin-1 was related significantly to lymph node metastasis and clinical stages,it may play a role on the promotion of tumor infiltration and metastasis,so it can be a reference in prognosis evaluation.2.The positive rate of EGFR was higher in primary lung cancer and precancerous lesion than normal lung tissue.It have become a new target for gene therapy.The positive rate of EGFR was significantly lower in SCLC than that in NSCLC, which suggested it can be a marker to distinguish SCLC and NSCLC.The positive rate of EGFR was related to lymph node metastasis and clinical stages,at the same time it may promote the malignant transformation and progression,so it may has a reference in prognosis evaluation.3.The positive rate ofβ-catenin was lower in primary lung cancer than that in normal lung tissue.The positive rate ofβ-catenin was significantly related to the degree of differentiation,clinical stages and lymph node metastasis,which suggested it can promote the malignant transformation and progression,so it has a reference in prognosis evaluation.4.There was significantly negative correlation between EGFR and Caveolin-1,β-catenin.There was significantly positive correlation between Caveolin-landβ-catenin.The study on the relationship of Caveolin-1,EGFR,β-catenin can reveal the mechanism related to lung cancer genesis and progress and provide theoretical evidence for gene therapy.5.Tissue microarray has the advantage of high throughput,high efficiency,saving reagent and time.It can reduce the experimental error and have prominent advantage in large scale research.
