Object: The paper is based on supramolecular chemistry "imprinting template" autonomous law of action and molecular structure topological technique,using molecular imprinting technique,to explain the characterization of guest component "imprinting template" of Lonicerae japonica flos(LJF)and Lonicerae flos(LF)in vivo and in vitro,which would provide experimental basis for solving the disputation between them.Method: The structure of LJF and LF guest component was determined by LC-QTOF-MS,and the 0-8th of molecular connectivity index(MCI)was calculated and correlated to their MCI to compare their "imprinting template" differences in vitro.LC-MS was used to determine the constituents of the LJF and LF in vivo.The topological structure of the blood components was calculated and compared the imprinting template.The metabolic dynamics of several major components were studied.The host and guest components were separated and synthesized to imprinted polymers to determine the difference of "imprinting template ".According to the first law of thermodynamics and Hess' law,a state mathematical model was established based on the burning of traditional Chinese medicine to determine the cold or heat.The anti-inflammatory effect and antibacterial activity were investigated to characterize the "imprinting template" feature in the LJF and LF.Result:1.The 390 metabolites with significant difference were determined in LJF and LF water extract by LC/MS-MS,and the topological index of 211 secondary metabolites was calculated,which was no significant difference by T test.There were differences between batches of topological index in 12 batches LJF and LF guest component.However,there was no significant difference "imprinting template" between the two groups.2.The 33 components were determined in rat blood samples,and the 7 components were determined,the metabolic processes were different.The result showed that there were some differences in the types and contents.however,there is no significant difference in the average MCI of the total components in the LJF and LF.3.The HPLC was used to determine the cross-adsorption of LJF and LF,which proved their imprinting effects were similar in vitro.The different polar guest molecule was used as template molecule to synthesis polymer.The results show that there is no significant difference in the selective adsorption of LJF and LF.4.According to the first law of thermodynamics and Hess' law,the heat produced by the organism should be the sum of the enthusiasm of drug and the organism before administration,minus the sum of the enthalpy after administration..The enthalpy of LJF and LF was determined with licoriceas the flat drug control,and the enthalpy was all negative,suggesting that the LJF and LF is cold.5.There was some inhibitory effect of 12 batches LJF and LF aqueous extract on Staphylococcus aureus and Escherichia coli.with a certain inhibitory effect on the infected mouse caused by aureus injection in the tail vein.The total MCI of blood components was0.526±1.7,0.607±0.87,respectively,which was no significant difference.6.The effect of LJF and LF on mouse ear swelling was found that the ear tumefaction inhibitory rate was 21.5% in the positive group,and 51.3%,51.6% in the high dose group of LJF and LF respectively,and there was no significant difference in IL-6 and IL-8 levels in vivo.The total average topological index was no significant difference in LJF and LF blood components by T test.The LJF and LF aqueous extract was swollen after rat was infected by carrageenan,to measure the paw volume of the rat,and calculate the swelling value and inhibition rate.Compared with the model group,the blank group and the positive drug group,the levels of IL-6 and IL-8 in the blood samples of rats were significantly different;There was no significant difference between the high,medium and low dose groups of LJF and LF,indicating that the anti-inflammatory “imprinting template”of LJF and LF may be similar.Conclusion: The in vitro and in vivo components and the content of LJF and LF were determined to be different by LC-MS.However,the average MCI of 0-8 order was similar,indicating that the two "imprint templates" were similar.The selective adsorption of LJF and LF aqueous extracts suggests that the imprinting behaviors were similar.According to the first law of thermodynamics and the state function of the action in traditional Chinese medicine and organisms,LJF and LF is cold.In combination with the classic antibacterial and anti-inflammatory model,the characteristics of supramolecular "imprinting template" in LJF and LF were similar,which was mainly composed of 5-7 C active space structure.
