Objective: Exploring the mechanism of compound Tinglizi decoction in improving airway remodeling in COPD-PAH rats based on classical pyroptosis pathway.Methods: Ninety SD rats were randomly divided into normal group,model group,low-dose compound Tinglizi decoction group,compound Tinglizi decoction medium-dose group,compound Tinglizi decoction high-dose group and western medicine group.ELISA was used to detect the expressions of IL-8,IL-6,IL-1 β,NF-βB and TNF-β in the bronchoalveolar lavage fluid of each group.HE staining was used to observe the pathological changes of lung tissue,and Masson staining was used to observe the changes of collagen fibers in lung airway tissue,RT-PCR and Western blot to detect the expression levels of pyroptosis-related indicators.NR8383 cells were treated with CSE and/or LPS,compound Tinglizi Decoction and Compound Tinglizi-containing serum for intervention.The activity of CCK-8 was detected,the cell pyroptosis was observed by transmission electron microscope,RT-PCR and Western blot to detect the expression levels of pyroptosis-related indicators.Results: 1.Compared with the normal group,m PAP,m CAP and RV/(LV+S)in the model group,compound Tinglizi decoction low-dose group,middle-dose group,and high-dose group were significantly increased(P<0.05),and the expressions of IL-8,IL-6,IL-1β,NF-κB and TNF-β in bronchoalveolar lavage fluid and TGF-β,NLRP3,caspase-1,GSDMD,IL-1β,IL-18 in lung tissue expression were up-regulated.Compared with the model group,the m PAP,m CAP and RV/(LV+S)in the low-dose group,middle-dose group and high-dose group of Compound Tinglizi Decoction were significantly decreased(P<0.05),and the expression of IL-8,IL-6,IL-1β,NF-βB and TNF-β in bronchoalveolar lavage fluid and TGF-β,NLRP3,caspase-1,GSDMD,IL-1β,IL-18 in lung tissue expression were down-regulated.2.20% CSE inhibited the activity of NR8383 cells most significantly.3.Compared with the control group,the cells treated with CSE,LPS and CSE+LPS had different degrees of activity inhibition and pyroptosis,and the CSE+LPS group had the most obvious cell activity inhibition and pyroptosis.4.Compared with the control group,the cells in the CSE+LPS group,the Compound Tinglizi Decoction-containing serum group and the Compound Tinglizi Decoction group had different degrees of cell activity inhibition and cell pyroptosis.Compound Tinglizi decoction group had the least inhibition of cell activity and cell pyroptosis.Conclusion: Our results show that Compound Tinglizi Decoction may improve airway remodeling in COPD-PAH rats by reducing the pyroptosis of NR8383 cells induced by CSE+LPS,and provide theoretical support for the clinical treatment of COPD-PAH with Compound Tinglizi Decoction.
