The sediment samples used in this study were collected from the east Pacific nodule province with water depth from 5000m to 5246m by cruises"DY105-12,14"which was organized by China Mineral Resources Association., during the time period from May 2001 to November 2003.Nodule province was compound including iron, manganese, nickel, Cobalt , and so on. These elements are valuable deep-sea source. China obtained the right to exploit in this area on 5 March, 1991.Microbiology are believed to play large roles in the metal cycling in many environments. Bacteria in this area possess potential study and applied value. To investigate the microbiology diversity , direct cultivation was used to estimate abundance and diversity of bacteria in samples. The sample analyses were carried out in the lab as following:(1) Sample treated by hot shock and sample untreated were used. Strain were isolated using 2216E, culturing iron bacteria medium, culturing manganese bacteria medium and other nutrient plates.(2) After microbial strains were isolated, growth patterns of 71strains were tested under a set of temperatures, salinities and pH values and enzyme production of the culturable microorganisms.(3) Microbiology diversity was analyzed based on 16S rRNA gene sequence. The main results are summarized below:Most isolates belong to cold-tolerance bacteria and can grow from 4℃to 30℃. Some alkaline-tolerance bacteria and salt-tolerance bacteria were also isolated from the sediment but no acid-tolerance. Some bacteria can degrade glutin, amylum, lipid...
