Gardenia jasminoides Ellis is a commonly-used traditional Chinese herbal medicine. And gardenia yellow pigment and geniposide are the two main chemical components in the fruit of gardenia jasminoides Ellis. Currently, the main use of gardenia is the extraction of gardenia yellow pigment. The exhausted solution of gardenia yellow pigment still contains a mass of geniposide. If it is discharged, it is not only pollution of the environment but also waste of the resources. Therefore, gardenia blue pigment was made from exhausted solution of gardenia yellow pigment and monosodium glutamate. And the text makes improvements to traditional craft. The study laid theoretical foundation for large-scale manufacture of gardenia blue pigment. The following are the several aspects of this work.1. The technology of immobilized enzyme was adopted, which is applied in the food industry. And the technology has replace the traditional technology of ferment in preparation of gardenia blue pigment. It will help to reduce the cost and improve the quality as a result of the characteristic of immobilized enzyme, which is repetition use and clear.2. The fermentation medium of aspergillus niger CMCC B 98003 is determined. It consists of 1.5% of bran,1.5% of rice bran,0.2% of (NH4)2SO4, 0.1% of KH2PO4,0.1% of CaCl2,0.05% of MgSO4·7H2O, and the rest is distilled water. The time of ferment cultivation is 84h.3. The immobilized enzyme is made by entrapment and the carrier is sodium alginate. The ripping method of immobilized enzyme is determined by experiment. First, mix up the enzyme solution with the sodium alginate solution of same volume. Second, extract the mixed solution by means of a syringe, and trickle the mixed solution into the 5% of CaCl2 solution. Last, put them into the safe of refrigerator till the pellets become white. The diameter of pellet is about 3.2mm.4. The conditions of hydrolysis equilibrium of geniposide were determined. The real-time monitoring of genipin was made by adding monosodium glutamate which is color development reagent. The experiments showed that this method is cogent and effective.5. Optimize the technological conditions of preparation of gardenia blue pigment. The preparation conditions of gardenia blue pigment showe that the dosage of monosodium glutamate is mgeniposide: mmonosodium glutamate=1:12, the response concentration is 2.4mg/mL which is concentration of geniposide in exhausted solution of gardenia yellow pigment, the pH of buffer solution was 7.4, the reaction temperature was 80℃.6. This text studied on the stability of gardenia blue pigment for related factors such as heat, light, metal ions, oxidants and reducing agent, acidic and alkaline. And the best conditions were determined, which are suitable for using and storing the gardenia blue pigment. The results show that the gardenia blue pigment's stability is better when the temperature under 60℃, in the opaque background, and the pH is at 6-8.
