Bioreaction with in-sute product removal is one key technique of biotechnology. But almost all of studies on microbial bioreaction with in-sute product removal were concerning on small molecules such as butanol and acetic acid, and researches on large biologic molecules must be founded in animal and plant cell culture . There were few bioreactions with in-sute product removal in studies on microbial fermentation producing large molecules such as gentamycin .Therefore, according to properties of gentamycin and biochemical and biological properties of cell culture, a new process of liquid-solid phase culture was developed for producing gentamycin in this paper. The chief studies include a method and mechanism of measurement of gentamycin concentration in microbial fermented broth by spectrophotometry, characters of ion-exchange resin absorping gentamycin and effects of ion-exchange resin to cell culture in process of liquid-solid phase culture of Micromonospora purpurea for producing gentamycin.l)The method and mechanism of measurement of gentamycin concentration in microbial fermented broth by spectrophotometry was investigated. The main method to determination of gentamycin concentration in the fermented broth was antibiotic microbiological method just like other antibiotics. That method was too complex to hold and would take much more time to be accomplished. By this new method, the determination of gentamycin concentration in the fermented broth was becoming simple, convenient and rapid . The effects of factors on accuracy of this method were also studied.2)The characters of Gentamicin adsorption on ion exchange resin was studied .The results show that the activity of Gentamicin didn't have any change after adsorded on ion exchange resin for five days and ion exchange resin that was whether sterilized or not hadn't any effact on the rule of Gentamicin adsorded on ion exchange resin. The kinetics model of Gentamicin adsorded on ion exchange resin was made and the effect of viscosity of Gentamicin fermentation liquor on the process of the simultaneous Gentamicin fermentation and product separation by ion exchange resin was discussed.3) In process of liquid-solid phase culture of Micromonospora purpurea, great effect on the growth of cells and the production of gentamicin for adding resin and penicillin wasstudied, and the optimum time and volume of adding resin and penicillin were also determined. The results show that the production of gentamicin in process of liquid-solid phase culture was 30% higher than that In process of liquid phase culture. On this condition, the new process of liquid-solid phase fermentation was investigated by adding resin and penicillin, the gentamicin production was improved 40% compared to the gentamicin production of liquid phase culture.4)By the end of the article, the kinetics model of process of liquid-solid phase culture of Micromonospora purpurea for producing gentamycin.was made. The results show that the kinetics model of process of liquid-solid phase culture of Micromonospora purpurea for producing gentamycin match the results of experiments.
