Study For The Technology Of Extraction, Purification And Quality Criteria Of The Bio-active Constituents From Bamboo Leaves

The goal of this experiment is to research a new second-class Chinese traditional drug .Methods for the determination and extraction as well as purification of the bio-active group,consisted of flavonoids and coumarin lactons from bamboo leaves are developed in this paper. At the same time,the quality criteria of the purified extraction of bamboo leaves(EBL) are made and the bamboo capsules are manufactured.The method for the determination of the flavonoids from bamboo leaves:The extracted solution of bamboo leaves is purified by the resin of polyamide first,then react with coloring agents NaNO2,A1(NO3)3 and NaOH. The concentration is determined by spectroscopy. Rutin is the control. The regression equations of the standard curves based on the concentration (C)versus the absorptive value(A) are:A=0.01264+0.01151(r=0.9999). The linear range is from 6.006ug/ml to 90.09ug/ml.The method for the determination of the coumarin lactons from bambooleaves is two-wave length spectroscopy. The determined wave length is 325.5nm ( A 1) and the control is 383.5nm ( A 2) . 7-hydroxide coumarin is the control. The regression equations of the standard curves based on the concentration(C) versus The difference absorptive value( A A) of the Al and A 2 are:AA=0.09107C+0.01150 (r=0.9986). The linear range is from 1.049ug/ml to 20.98ug/ml.Take the contents of the flavonoids and the coumarin lactons as reviewing targets,by good single factor selecting experiment and Orthogonal Design,the best technology for extracting bamboo leaves is established. It consists of 55%ethanol as solvent,!00 as extracting temperature,three times,two hours per time and the solvent volume is ten times more than the weight of the bamboo leaves at the first time,seven times at the second time and four times at the third time. The content of the flavonoids in the bamboo leaves is about 1 Omg/g and the coumarin lactons is about 7mg/g.The technology for purifying the EBL is established by the combinatory using of two types macroporous absorption resin HPD600 and D101. After the purification,total content of the flavonoids and coumarin lactons is more than 50% .The quality criteria of the purified EBL are pilot drawn. Identify the constituents by chemical reactions and thin layer chromatography. Check up some items such as drying weightlessness,burning residue,arsenic salt,heavy metal,residue of organic solvent etc.The purified EBL as material drug,microlite cellulose as accessory,the bamboo capsules are manufactured after making the particles. The ratio of the two parts is one to one and the wetting agent is 80% ethanol. The load quantum of the capsule is 250mg.The stability of the capsules is pilot studied by high temperature,high humidity and strong light irradiation experiments.
