Phenol is one of main pollutant of industrial waste water in these days.The researches for phenol transaction come out continuously all around the world, biodergradation is the useful ,up-and-coming technique of all. But there are many heavy matal salts in the waste water, its restrain for biodegradation of microorganisms is strong, and that greatly restricts biodegradation used in phenol-contaminated water. R.metallidurans CH34 is just the phenol-degrading wanted strain with anti-heavy metal feature.This paper examined its characteristics of growth and phenol degradation.Immobilization resolve a series of matter that it is low-efficiency and hard to control for free cell in biodegradation. Many people try to apply immobilization to phenol degradation, and it work well. In this paper R.metallidurans CH34 with anti-heavy metal feature was immobilized, the mithod of immobilization and conditions of operation was optimized, and its results were test.l.The basic parameters of physiological characters for R.metallidurans CH34 were determined. Before a series of characteristics are further examined, it was necessary for R.metallidurans CH34 to master its basic characteristics and growth conditions,for example choosing medium, growth curve and a series of cultivating conditions(temperature,pH,OD). The results :Using M9 abio-medium;Working out growth curve;Optimal temperature,pH and liquid capacity are 30℃, 7and20%(v/v) respectively.2.Domestication and characteristic of phenol biodegradation of R.metallidurans CH34. Study the effect of domestication of R.metallidurans CH34 which is cultivated by different means:solid medium and liquid medium, it shows that domestication can improve the characteristic of phenol degradation. A series of characteristics of phenol degradation are studied to be ready for immobilization, and it is refer to the characteristics of immobilization.3.Preparation of immobilized cell of R.metallidurans CH34. At first SA, PVA and their mixed carriers are chosen for beginning carriers; After comparation furthely SA is chosen for carriers; In the other hand the conditions of simulated waste water are decided to use; The optimal conditions for immobilizing cells were obtained with cross act experiment: quantity of damp cell 4.8g/L, CacL2 concentration%, SA concentration 5%. Intuitionistic analysis and square difference analysis of cross act experiment showed that CacL2 concentration was the most important factor for immobilization. Kinetic simply result of analysis of cross act experiment was as same as the result which was obtained as the optimal one in the cross act experiment. At the same time adsorption capability of SA carriers for phenol was studied.4. The capability of immobilized cell of R.metallidurans CH34. In the hand of phenol degradation,immobilized cell improved its capability of degrading phenol greatly; It put up good active during the continuously phase of degrading phenol, The upper limit of phenoldegradation had improved from 10mM to 15mM and the rate of phenol degrading had also been greatly improved; The surface of carriers of immobilized cell cultivated was observed using electron microscope,accordingly it embodied the growth situation of strain in the carriers which was cultivated for a period of time, It just was corresponding to the situation of phenol degradation. The immobilized cell had better resistance to heavy matal and degraded phenol faster than free cell. When immobilized R. metallidurans CH34 degraded phenol.it had greatly improved resistance to the interruption of additional carbon resources.
