It has become fucosCarotenoid is used as the natural pigment and health food composition. For increasing agricultural product additional value and providing theories base for establishing standard, this paper have been studied as follow.The extraction process of Maize Yellow Pigment form Corn Gluten Meal has been studied and the appropriate extract condition of direct extraction and the extraction after enzyme-hydrolysis was confirmed. It has been found that the appropriate solvent of direct extraction was acetone: petroleum ether=l:l(v/v). The purity of Maize Yellow Pigment was elevated effectively. The method of enzyme-hydrolysis has been studied by single factor experiments and orthogonal test. The results showed that the optimum parameters as follow: enzyme concentration 1.4%, substrate concentration 5% and duration of hydrolysis 6h at 40℃, pH7 by neutral proteinase. The extraction effect was highly improved with the method of enzyme-hydrolysis that the product ratio was increased 126.8%. The analysis method of carotenoids by thin-layer chromatography (TLC) and high performance liquid chromatography (HPLC) was studied in this paper. Silica gel G as absorbent. The carotene and the Xanthophyll was analyzed by Acetone : petroleum ether (1:100, v/v) and Acetone : petroleum ether (3.5:6.5, v/v) as eluent, the mixture and unknown were detected by two conditions. The carotenoids were separated by HPLC with Diamonsil? C18 column and a mobile phase of acetonitrile and ethyl acetate with the following gradient eluention were used. The method did not appear away. Established the handles method of carotenoids in health food. The different methods and solvents have been comfired, including meal supplement and tomato beverage. Carotenoids of seven meal supplements and five tomato beverages were analyzed qualitatively and quantitatively. It was found that carotenoids were inccording with identifiers for meal supplements, but accroding with tomato beverages. The TLC seperation condition of four kinds of carotenoids and Sudan has been confirmed. This research established the academic foundation to set down correspond standard.
