Planar gel electrophoresis is an indispensable tool for the separation and analysis of DNA, RNA and protein; thin layer scanning chromatography (TLSC) is a main method for the separation and analysis of organics, medicine, especially the natural medicine, TLSC is also one of the main techniques for the fingerprint of Chinese traditional medicine. Biochip is an advanced technology for gene sequence determination, gene expression, gene diagnosis, protein function research, high through-put for candidate medicine. The signal-extraction principle of this entire analytical technique is 2-dimensional image detection. So computer -associated digital image processing can be utilized.Therefore in this thesis, a new software for computer-associated digital image processing was developed by using VC++, Delphi language. It was expected to has such following functions:1) Use cheap common scanner or digital camera /ultraviolet analyzer to replace expensive professional thin layer scanning chromatography (TLSC) and the planar gel electrophoresis imaging system.2) Be able to do qualitative and quantitative analysis of TLSC and planar gel electrophoresis.3) Be able to calculate and compare the similarity of chromatography of Chinese traditional medicine's fingerprint.4) Be able to detect 2-dimension array imaging, laying a foundation for biochip detection.A number of applications have shown that this digital image processing system which combines the software with a common scanner or a digital camera / ultraviolet analyzer has such following functions:1. Be able to do the qualitative and quantitative analysis of TLSC and planar gel electrophoresis:1) Give the value of Rf, a qualitative parameter in TLSC.2) Be able to transform quantitatively a spot or a band (visible or fluorescent) on a planar of TLSC or planar gel electrophoresis into digital value.2. Be able to calculate and compare the similarity of Chinese traditional medicine's fingerprint:1) The file format of fingerprint can be windows general formats such as BMP, JPG and GIF. The internet file format can also be utilized.2) Fingerprint on record paper can also be utilized.3. In the aspect of array detection1) A visible array of 6400dots/cm2, with an area of each dot less than 0.015mm2 can be clearly discriminated.2) A fluorescent array of 64dots/cm2, drawn with a lactofalvin solution of 1ppm, each dot containing 1ng lactofalvin, can be clearly discriminated.4. Having friendly humane-computer interface in the software, database technique was applied, so it permits data auto list, auto edition, and saving a file as Excel format.Compared with professional instruments, the proposed digital image processing system has the advantage of simple equipment, lower price and convenient operation, so it is easy to spread. This system provides an effective and economic way for planar chromatography research and application, and for quality evaluation and control of Chinese traditional medicine in many common laboratories of colleges and small-middle enterprises in our country.
