Bamboo bark, a traditional Chinese herb called Zhuru, is approved as 'material that can be used in health foods by Ministry of Health, P. R. China. However, few published studies systematically reported the active components and biological activities of bamboo bark both in China and other countries. In the present study, the chemical composition of the extract from bamboo bark obtained via supercritical fluid extraction (SFE) technique (named as EZR2002) and the safety evaluation of EZR2002 were systematically discussed. The biological activities of EZR2002 on regulating blood lipid, lowering blood pressure and anti-fatigue were also demonstrated in our study. The main results were described as follows:1 , EZR2002 was obtained from the coarse powder of bamboo bark after dynamic supercritical CO2 extraction (25h) under the condition of 50-65℃, 25-35 Mpa with a general yield of 2%.2, EZR2002 is a yellow or green-yellow powder with the melting point from 74℃ to 79℃. Its IR spectrogram performed by potassium bromide showed characteristic absorption peaks in the wavelength of 2917, 2849, 1716, 1463, 1382 and 720cm-1, respectively. Its UV spectrogram dissolved by spectroscopic pure CH2Cl2 with a scan range from 300 to 700nm showed a strong absorption in the wavelength of 412nm, a sub-strong absorption in the wavelength of 665nm and a weak absorption in the wavelength of 505, 535 and 605nm. EZR2002 mainly contains a group of pentacyclic triterpenoids, including friedelin, lupenone and lupeol detected by GC-MS. Total triterpenoids content was (55-75) % measured by vanillin and perchloric acid colorimetry using friedelin as standard, while the content of friedelin and lupenone were (13.3—15.2)% and (1.3 1.5)% quantitatively analyzed by GC using friedelin and lupenone as standard, respectively, in different batches of EZR2002.3, Acute toxicity studies demonstrated that the oral LD50 values of EZR2002 were more than 10 g/kg body weights both in rats and in mice which belonged to actually no toxicity according to the criteria of acute toxic classifications. It had no mutagenicity judged by negative experimental results of Ames test, mouse bone marrow cell micronucleus test and mouse sperm abnormality test. As for the 30 days feeding study in rats, no abnormal symptoms or deaths had been found in each group of rats during the experiment. No significant difference had been found in body weight, food consumption and food availability of rats in each test group compared to the control group (p>0.05). Furthermore, no significant difference had been found in corresponding hematology indexes, clinical chemistry indexes and organ/body weight ratio compared to the control group, either (p>0.05).
