Continuous rod ion-exchange chromatographic columns have been widely Used in separation and purification of protein, peptide, nucleic acid, polyribonucleotide, and polysaccharide in recent years due to their excellent characteristics such as 1). open ended structure, allowing macromolecules to distribute and pass through quickly, which will make rapid separation and preparation possible; 2). porosity, high surface area and exchange capacity; 3). hydrophilicity, which would not result in the inactivation of protein and enzyme, for their weak adsorption of bio-macromolecules. Furthermore, they can also be used as the materials of solid phase extraction (SPE) in the pre-concentration of trace components before analysis. They are safe, convenient and effective, compared with conventional enrichment techniques, and automatic on-line analysis without overfull consumption of organic solvent becomes a reality.In this paper, continuous rod ion-exchange chromatographic columns were prepared by in-situ polymerization within a confines of 64 X 4.6 mm I.D.. Their adsorption characteristics of bio-macromolecules and on-line enrichment of anions were investigated. There are three chapters in this paper.Chapter one: Review of the recent advances in the stationary phase of ion-exchange chromatography (IEC), including inorganic, organic and composite matrices, and the applications of IEC in biochemistry was introduced.Chapter two: A continuous rod chromatographic column with glycidyl methacrylate as the functional monomer and ethylene dimethacrylate as the cross linker was prepared, and carboxyl groups were bonded on the column by on-line modification. To study the characteristics of the chromatographic column, three factors, such as flow rate, temperature and pH, were investigated with Lysozyme and bovine serum albumin. The results indicated that the column could adsorb basic protein obviously, and had typical characteristics of weak cation-exchange chromatographic column. It could be used for purification of basic protein, and its reproducibility and stability were both perfect.Chapter three: The analysis of trace thiocyanate and iodide by means of on-line enrichment technique was presented in this article.
