Grape seeds contain quantitative grape seeds oil (GSO) and procyanidins (PCs).GSO is rich on unsaturated fatty acids such as linoleic acid;widely distributed in thekingdom of plants, PCs are kinds of polyphenone compounds with strong bioactivitiesand pharmaceutical effects of anti-oxidant activities, which can be applied in medical,health care, and cosmetic industries. It is one of the most popular herbal medicines inthe world. Consequently, it must be realistic and meaningful to seek an appropriatemethod to extract GSO and PCs from grape seeds.In this thesis, the integrate technology of supercritical fluid extraction (SFE)process and macroporous adsorption resin process for extracting GSO and PCs fromgrape seeds was studied on the basis of the previous research results obtained. Inaddition, the other methods extracting PCs from grape seeds were compared.The process of extracting GSO from grape seeds was firstly studied using a 5LSC-CO2 extraction plant. The yield of GSO in this experiment was found to be12.85% average from 3 times.In order to improve the quality of PCs and to reduce the content of waterinsoluble substances in PCs to the level of less than 5%, the integrate technology ofwater solution deposited by alcohol and macroporous adsorption resin was employed.The influences of some factors on both the yield and content of the PCs extractedfrom grape seeds were investigated in the process of extraction, separation andpurification. In the sections of extraction and separation, the concentration and theflow rate of the alcohol were investigated. In the further research of purificationprocess with macroporous adsorption resin, five kinds of macroporous adsorptionresin with different polarity were used for adsorbing the PCs, in which the 1# resinwas considered to be the best. Then the effects of eight factors on both the adsorptionratio and content of PCs were explored in dynamic adsorption process, which includethe flow speed and amount of the adsorbate solution, the concentration of adsorbatesolution, the inner diameter of the adsorbing column, the operation temperature, pHvalue of the adsorbate solution, the flow speed and the concentration of desorbingagents. As a result, a reasonable conditions of adsorption and desorption wereobtained. The experimental results show that, under the optimum conditions in theprocess of extraction, separation and purification, the PCs with high content 97.82%,high yield of 4.88% and insoluble matter yield in water of 1.67% can be obtained,which is better than the traditional methods.Based on the above studies, five kinds of technological processes for extracting,separating and purifying the PCs from grape seeds were further compared.
