Environmental hormones, also called environmental endocrine disruption chemicals, act as estrogens in human and animal bodies. Most of them are discharged into the environment with the production and the life of the humanity. It has been found that environmental hormones existing in the environment have adverse effects on endocrine systems in human and animals and lead to reproductive, developmental and behavioral disorder. In recent years, environmental hormones have become the third environmental problem after the global warming and ozonosphere destruction. Now the research on the screening and determing the environmental hormones has become one of the hotspots of environmental science. Two new methods were researched in this paper: immunofluorescence method and flow injection chemiluminescence method, bisphenol A (BPA) , 2,4,6-trichlorophenol(TCP) and p-tert-butylphenol were determined respectively. The determination of these three environmental hormones could achieve comparatively perfect results by using these two methods.Immunofluorescence method was developed for the analysis of BPA and TCP. First, the haptens of BPA and TCP were synthesized. Then thehaptens were conjugated to protein BSA to prepare antigens. The conjugates were identified by the method of ultraviolet. The result demonstrated that the conjugates were synthesized successfully. The antibodies were obtained after three months by injecting the immunogen in rabbits. Using the purified antiserum of highest specificity, an antigen coated immunofluorescence method and a direct competitive immunofluorescence method were developed. With the antigen coated immunofluorescence method, BPA was determined in the drinking water and river water. The linear calibration range was 10"7~10"4mg/mL, the detection limit IC20 was 3xlO~7mg/mL and the recoveries were between 96%~108%. The result was satisfactory. With the direct competitive immunofluorescence method, TCP was determined in the tap water and river water. The linear calibration range was 10'6~5xl0"4mg/mL, the detection limit IC2o was 2.4xlO~6mg/mL and the recoveries were between 95%~106%. The result was satisfactory. The chemiluminescence behavior of Luminol-H2O2 and p-tert-butylphenol in alkaline solution was researched by flow injection analysis. The related factors affecting chemiluminescence intensity were optimized. The method for determination of p-tert-butylphenol was established by flow-injection-chemiluminescence. The detection limit obtained was 4xlO"7 mg/mL. The linear range was 2*10'6~ lxlO'4 mg/mL. The relative standard deviation of 1.45% was obtained by 11parallel determinations of 10"5 mg/mL p-tert-butylphenol. This method was satisfactory for the determination of p-tert-butylphenol in water.Wang Lan (environmental science and engineering) Supervised by professor Zhuang Huisheng...
