This paper includes two parts. Firstly, we synthesized a series of porphyrin-5-Fluorouracil and metalloporphyrin-5-Fluorouracil compounds for anticancer drugs;secondly, the study was preceded on the supercritical CO2 fluid extraction technique of the efficiency component from Podophyllum emodi Wall.5-Fluorouracil is a kind of popular antimetabolic drug used in clinic. It has the similar structure with uracil which is necessary to the metabolism of cancer cell. It contents with uracil in the same enzyme system, blocks the metabolic link, impedes the synthesis of DNA, and then prohibits the propagation of the cancer cells. As a kind of antitumor drug, 5-Fluorouracil has inhibition effect on many kinds of cancer, but it also seriously hurts the normal cells while it kills the cancer cells.It is known, porphyrin and metalloporphyrins have special affinity to cancer cell because their distinctive structure. They can be taken up and retained by tumor tissue selectively. In order to decrease the toxicity of 5-Fluorouracil, a series of porphyrin-5-Fluorouracil and metalloporphyrin-5-Fluorouracil compounds were be synthesized. We expected these compounds can reach tumor tissue and kill cancer cell directly. The further work is in progress.Supercritical Fluid Extraction (SFE) is a new technique developed lastly for chemicals extraction and separation. SFE with subsequent HPLC analysis was utilized to extract activity components such as podophyllotoxin, deoxypodophyllotoxin, kaempferol and quercetin from the roots and stems of Podophyllum emodi Wall. The extraction yields obtained by SFE were compared to those obtained by organic solvent extraction and were found to be comparable or lower. In addition, we used the orthogonal experiment which conducts with 3 factors and 4 levers in order to find the best dynamic extraction conditions.
