In this paper, an accurate analysis method for the trace tetracycline antibiotics(TCs), such as oxytetracycline, tetracycline, chlortetracycline in animal tissue was investigated systematically in order to improve the traditional techniques. The pretreatment procedure of animal tissue samples was also discussed, and the accurate HPLC-ESI/MS/MS and HPLC-UVD methods that simultaneously determinate oxytetracycline, tetracycline and chlortetracycline were set up.During the pretreatment procedure, the extraction of the three TCs was carried out by liquid-solid extraction, and solid-phase extraction (SPE) with C18 column was used to clean up the extract. The optimum pretreatment conditions were obtained by optimizing the conditions of the extraction and cleaning up. Average recoveries obtained by this pretreatment procedure ranged from 83.9% to 93.7%, and the repeated relative standard deviation (RSD) for the recovery ranged from 2.4% to 2.9%.The conditions of HPLC-ESI/MS/MS and HPLC-UVD were developed for simultaneous determination of three tetracycline antibiotics in animal tissue samples. Under those conditions, these three TCs could be well separated, and the repeated RSDs of HPLC-ESI/MS/MS and HPLC-UVD for the analysis TCs mixed criterion sample were 0.95.0% and 0.70.9%.
