The extraction and purification of solanesol from discarded tobacco leaves were studied.The process includes solanesol extraction, rude extracts saponification,and rude solanesol purification.First , a more accurate determination method of free solanesol in discarded tobacco leaves was found.Content of free solanesol in various tobacco leaves was determinated with this method.The results showed: Free solanesol content in discarded tobacco leaves was only slightly lower than in the first-class tobacco leaves,The tobacco peduncles also had value in use. At the same time,Straight-phase HLPC was used for quantitative determination of the free solanesol content and TLC was found for fast qualitative analysis of solanesol.Secondly,it was investigated that use of supercritical carbon dioxide fluid extraction process and organic solvents extraction process for the extraction of solanesol in discarded tobacco leaves. On the basis of Single-factor experiments inspection of the respective main factors influencing extraction process, the respective optimal extraction conditions were obtained by orthogonal optimize design. Under their respective optimal extraction conditions,the extraction yield of solanesol with 1.28% was obtained in 92.1% yield by supercritical carbon dioxide fluid extraction method and the extraction yield of solanesol with 1.25% in 90% yield by organic solvent extraction method. The results showed: compared with organic solvents extraction method:,Supercritical carbon dioxide fluid extraction method had the advantage of less solvent consumption, shorter extraction time and higher extraction yield.It is more suited to extract solanesol from discarded tobacco leaves. Saponification process of rude extracts was established. Saponification solution via bothing, neutralization with diluent hydrochloric acid and delamination.The solanesol with 52.6% purity obtained.Finally,two kinds of purification technics for rude solanesol were investigated.Rude solanesol via adsorption by column chromatography silica gel of 100-200 mesh, gradient elution with the different concentrations of co-solvents acetone-light petroleum, with acetone,and vacuum drying.The purity of solanesol was increased from 52.6% to 93.5%, reaching the pharmaceutical purity requirements. This method had character of theoperation simple, the purity of product high. Fractional recrystallization method, as a new purification technics,was explored to refine solanesol from rude solanesol.By fractional recrystallization in turn with acetone, methanol and acetonitrile,and vacuum drying.
