| This paper reviewed the biological and ecological characteristics of the puna chicory (Cichorium intybus L). The puna chicory root was rich in inulin. Inulin has been widely used as a functional food ingredient in European and American. The inulin market is booming in China recently. The advances in the research of inulin were reviewed too,including the sources,properties and physiological functions of inulin,the application in food industry and the evaluation of safety. The manufacture conditions in the domestic and foreign countries were analyzed as well as the application. The development of chicory inulin must have a extensive prospect.There is no systemic report on the research of extraction and purification technique of chicory inulin in china.The objective of this paper is to lay a foundation for compositive utilization and exploitation of chicory. On the bases of previous research of inulin and the facilities of the laboratory, this thesis systematically studied a colorimetric method for quantitative analysis of inulin, optimized the parameters of extraction using orthogonal design and the parameters of decolor using active-carbon by orthogonal design. The process of deprotein of crude inulin was further researched. The major results were shown as follows:The content of inulin was detemined by UV-VISspectrophotometry after it has been colored. Using dextrose as a standard substance, 1 ml resolution of inulin diluted 500 times and mixed 5 ml anthracene ketone reagent, heating for 15min in boiling water, and shaded for 15 min, determined concentration of total sugar, assured detective wavelength of total sugar was 615 nm. Using dextrose as a standard substance and 3,5-Dinitrosalicylic acid assured detective concentration of reduced sugar in the crude inulin. These methods is rapid ,accurate and repeatable. The effects of extraction time, extraction temperature and material to water ratio on the yield of inulin were investigated. The optimal condition for the extraction of inulin polysaccharide was established by orthogonal experiment. The optimal parameters were: temperature 85℃, water ratio 1:30 and time 60min. The yield of inulin from Puna chicory (Cichorium intybus L.)root could achieved 57.84% under the condition established. |
