Inulin is composed of different polymerization degree fructans.It is a natural functional food polysaccharide which has the dual effect of dietary fiber and bifidus factor.And inulin has been approved as a new resource for food by China's Ministry of Health in 2009.Due to the Jerusalem artichoke(Helianthus tuberosus L.)has many advantages(rich source,wide adaptability,strong resistance,low cost,etc.)in our country;it has been listed as inulin preferred raw material production by National Ministry of Science and Technology and the Ministry of Agriculture.This thesis took "Helianthus tuberosus L.Ht1" as the raw material,systematically studied and discussed the extraction and purification technology of Jerusalem artichoke inulin.And made a test run of Jerusalem artichoke inulin pilot production line in Saline Land Agricultural Science and Technology Co.,Ltd.,which located in Dafeng,Jiangsu Province.The main results are summarized as follows:1.With phenol-sulfuric acid colorimetry to determine the total sugar,DNS colorimetry determination of reducing sugar,the inulin content is total sugar content minus reducing sugar content.Through verification test,stability test,repeatability test and precision test proved that microplate reader trace method was measured with a good repeatability,fineprecision,simple and rapid operation,and less sample dosage.2.The best process of hot water single extraction is when the ratio of material to waterl:15,extraction 60min under 85 ?.At this time,the total sugar yield was 70.22%,reducing sugar was 5.60%,and the yield of inulin was 64.62%.The influence extent to inulin extraction rate is the extraction temperature,the ratio of material to water,extraction time in a descending order.If extraction in several times,the preferred process is:in the first time,the ratio of material to water 1:10,extract 40min;in the second time,the ratio of material to water 1:5,extract 20min(the extraction temperature was 85 ?).At this time,the extraction rate of total sugar is 71.76%,the extraction rate of reducing sugar is 4.92%,and the total inulin extraction rate is 66.84%3.Using activated carbonon inulinextract for the first time decolorization,single factor-orthogonal experiments show that the influence extent to decolorization ratio is activated carbon dosage,decolorization time,decolorization temperature in a descending order;the influence extent to inulin retention rate is activated carbon dosage,decolorization temperature,decolorization time in a descending order.Decolorization rate and inulin retention rate of 50-50 weight calculation decolorization effect;the best decolorization process is carbon dosage 5.0g/100mL,decoloring 40min under 35 ?.At this time,the decolorization rate is 80.25%;the inulin retention rate is 92.39%.4.Desalination experiments comparing the desalination effects of resins PK228?D001 and D113.Results show that the influence extent to desalination rate is PK228,D113,PK228 in a descending order.But PK228 lead pH of inulin extract to acid.The influence extent of regeneration resin desalination rate is D001,D113,PK228 in a descending order.At this time,PK228 has no influence on pH of inulin,but its desalination rate is the lowest.Therefore,take the cost and desalting effect into account,select resin D001 for desalination.5.Decoloration experiments comparing the decoloration effects of resins D201?D301 and PA312.Results show that the best decolorization resin is PA312,D201 is slightly inferior.But PA312 is quite expensive,and its decolorization rate is only a little higher than D201.So choose D201 for the second time decolorization.The single factor-orthogonal experiments show that the influence extent to decolorization ratio is D201 dosage,decolorization time,decolorization temperature in a descending order;the influence extent to inulin retention rateis decolorization time,D201 dosage,decolorization temperature in a descending order.Decolorization rate and inulin retention rate of 50-50 weight calculation decolorization effect,the best process of secondary decolorization is D201 dosage 9.0g/50mL,decoloring 30min under 25 ?.At this time,the decolorization rate is 87.29%,the inulin retention rate is 88.36%.6.The nanofiltration experiment results show that two different type nanofiltration membranes have a certain effect on inulin extract concentration,MWCO 1000 and MWCO 2500 can intercept FOS,but MWCO 1000 cannot separate saccharose.So use MWCO 2500 to purify and concentrate the inulin extract.More filter make higher purity,but the recycle of sugar in dialysis fluid will be more difficult.7.Inulin made in lab test is pure white,the total sugar content in1.0g crude Jerusalem artichoke powder is 0.9080g,the inulin content is 0.8064g,and inulin purity is about 88.81%.And inulin made in pilot scale test is mike white,the total sugar content in 1.0g crude Jerusalem artichoke powder is 0.8031g,the inulin contentis 0.6649g,and inulin purity is about 82.79%.
