Possessed of particular structure, collagen has low antigencity associated with high biocompatibility and constitutes one of the major raw materials for biomaterial applications, which is being much more important in biomedical realm.The pigskin, which is a kind of rich, cheap and reproducible resource, has many advantages in component and structure, but neglected as raw material of collagen extraction. Based on the acquired results of pigskin's purification, the techniques of collagen developed with pepsin were optimized, and the spray drying parameters were chosen. Biomedical collagen powder's physicochemical properties were characterized and applied in collagen-based membrane, and the powder was evaluated in vivo.The main results of the research go as follows:1. The results of orthogonal experiments and SDS-PAGE electrophoresis showed the optimized techniques were 4℃, pH 2.2, the usage of pepsin was 2% and the ratio of liquid to solid matter was 50.2. The extraction process could be simulated by first order Hill equation and conformed to the features of Michaelis equation, and the change of hydrolyzation velocity could be fit by first order exponent decay equation. The equations confirmed that the pepsin usage of 2.0% and the hydrolyzation time of 8h were reasonable. The time necessary for hydrolyzation velocity decayed from the greatest to the half was about 2.59 h±0.19 h.
