Crystallized mother liquor of glucose was analyzed by high performance liquid chromatography (HPLC) on an ion-exchange column. The results indicate that mother liquorâ… contains over 45g/100ml sugar compositions, and the purity of glucose is over 85 %.In order to extract glucose from its crystallized mother liquor, a special kind of cation-exchange resin, which has the property to adsorb glucose strongly while the other impurities were weakly retentated was synthesized by us. The resin was prepared using disperse polymerization method based on the structure of poly (styrene-divinylbenzene) with crosslinker and sulphonic acid as functional group and Ca2+ as the inverse ion. The influences of synthesized conditions on the diameters and size distributions of the microspheres were investigated by orthogonal_designing method. Meanwhile the microspheres were packed into analytical columns to test their adsorption capacities on glucose. The results proved that this kind of adsorbent was suitable for separating glucose from the mother liquid.The optimum operation conditions, such as operation temperature, the rate of length and diameter of the column, the amount of feed and the flowrate of the eluent) of a preparative column packed with the Ca2+ resin were investigated. It showed that under the optimum condition, the recovery of glucose was over 84% and the purity is more than 95%.The oligosaccharides were found in the crystallized mother liquor of glucose as well. HPLC is used to analyze the composition on an ion-exchange column and a NH2-bonded carbohydrate column, respectively. The results indicate that mother liquorâ…¡contains over 4g/100ml sugar compositions, and the purity of isomaltose is over 37%.In order to separate isomeric compounds from functional oligosacchardes, the packing materials which used in chromatographic industries were prepared by the modification functional group NH2 bonded on silica gel. The influences of preparation conditions on NH2 amount and the separation effects were investigated by orthogonal designing methods. The bonded phases obtained were characterized by infrared spectrum, elemental analysis and the chromatographic performance. The results showed that the stationary phase had excellent chromatographic properties and resistance to hydrolysis between pH 2~12. It could be used for the efficient separation of isomeric compounds from oligosacchardes.Maltose and isomaltose were separated by preparative columns packed with the NH2-bonded self-produced resins. Under the optimum conditions the recovery 82% of isomaltose (purity over 97%) has been achieved.
