The saponins is one of the mainly active ingredients from Momordica charantia.L . It is reported that the saponins from Momordica charantia.L have been used for the treatment of hyperglycemia, high blood-fat, tumor, AIDS, antiphloggistic, immune function to human being, etc. In this thesis, our research focuses on extraction, fractionation and isolation of saponins from Momordica charantia.L , and we also make a study of the function of inhibiting microbe.First, the powder of Momordica charantia.L is analyzed. The result show that the powder content moisture 11.20%, ash 2.52%, crude protein 22.72%, crude fat 7.41%, saccharides 43.55%,others 12.59%.In the next step we firstly used a novel method, column extraction process to extract saponins from Momordica charantia.L, which was compared to the conventional reflux extraction methods in terms of the yield, the ratio of raw material to ethanol. For comparison of the two techniques, both at the base of orthogonal experiments, all extraction parameters were kept the optimum conditions, respectively. Results showed that the column extraction process compared with the conventional reflux extraction methods to increase the yield from 3.36% to 4.13%, suggesting it can be alternative technique to the conventional reflux extraction methods. Then, we optimize the extraction condition again by SAS and Matlab analysis software again. At last, the rude saponins extration is accounted for 4.30% of the dried power of Momordica charantia.L.Eight kinds of macroporous resins were selected to compare their absorption and desorption of saponins. Result showed that ADS-7 resins was the optional to purify the saponins from Momordica charantia L. And the ADS-7 resin's dynamic absorbent capacity was also studies. Then we used silica gel column chromatography to purify the saponins, and make a study on the technics. The experiment result showed that the purity of saponins were 93.2% respectively, and recovery ratio was 88.9%.The inhibition effect of saponins from Momordica charantia.L on microorganisms and the effect of temperature, concentration of NaCl, pH value and ultraviolet radiation on its antibacterial activities were studied by the method of agar diffusion. The results showed: the saponins had stronger inhibition effects on on some G+ and G- bacteria .The antibacterial activity of saponins were stable under heating and ultraviolet radiation, concentration of NaCl, but the antibacterial effects is not very well at pH4~5.
