Enantiomers often present different biological activities for their different pharmacokinetic, pharmacological or toxicological properties. Thymopentin consisting of five peptides has 31 kinds of enantiomers. For drug development, quality control and researching relationship between activities and their molecular structures, it is essential to lucubrate the chiral separation method for Thymopentin's enantiomers and provide a exact, simply and quick analytical technique to attain adequeat resolution of 31 kinds of Thymopentin's enantiomers.In this thesis, chiral separation for Thymopentin's enantiomers was investigated using separation technique chiral mobile phase additive(CMPA) by CE techniques. The paper consists of the following contents.First of all, separation methods of pairs'just having one chiral central were investigated. Pairs were separated by using three kinds of chiral selectors respectively, includingβ-cyclodextrin (β-CD), hydroxypropyl-β-cyclodextrin (HP-β-CD) and 2, 6-dimethyl-β-cyclodextrin (DM-β-CD). After analysing the results, the separation orderliness was discussed, just as following. First, When chiral central position is different, so is chiral selectivity, it's seriation is Tyr>Val>Lys>Arg>Asp. Second, for cyclodextrin kind, it is appropriate to use HP-β-CD when chiral central at Arg or Lys. It is appropriate to useβ-CD when chiral central at Asp. It is appropriate to use DM-β-CD when chiral central at Val or Tyr. Under such background, a mechanistic study of enantiomeric separation was discussed.Besides, Thymopentin's enantiomers were separated when using DM-β-CD and sequencely found 10 enantiomers could be splited under this condition. Then several chromatographic conditions were studied step by step and finally the optimal condition was attained.
