In this paper, the extraction, separation and purification of alkaloids from Sophora alopecuroides L were investigated. The main results were as following:1. The content of the total alkaloids in Sophora alopecuroides L is analyzed by acetic acid buffer solution which pH was 5.6.and the result is 8.70%.2. The optimal parameters of extraction were obtained ultrasonic assistant experiments and orthogonal tests: 65% ethanol,pH 0.5 hydrochloric acid, ultrasonic frequency 28 kHz, ultrasonic time 20min, According to the analysis of extract, the total amount of alkaloids reaches to 21.8% in total solid substance. The extraction ratio was 75.76%.3. Studied the method of separating Alkaloid in Sophora alopecuroides L.,The silica gel preparation column was used to separate and purify the monomer of alkaloids and three monomers of alkaloids are separated and then crystallized. In this paper, we deeply studied the each parameter of purify matrine and oxymatrine. TLC, ultraviolet spectrum were used to identify the qualification of crystallization, and and high performance liquid chromatography (HPLC) was used to identify the purification of crystallization. In this paper, we found out ideal method of which got high purification matrine and oxymatrine from. Sophora alopecuroides L. Purification exceeded these alkaloids were identificated as oxymatrine, oxysophocarpine and sophoridine. The purity of them was analyzed by HPLC and was 98% respectively.4. By studying cation exchange resin , determined the optimum parameter of extraction technology of Alkaloid in Sophora alopecuroides L.Antimicrobial activity of Alkaloid in Sophora alopecuroides L. were tested against bacterium. Results showed there had strong antimicrobial effects for bacterium.
