| Sophora alopecuroides L., a leguminous plant of Sophoral, is widely distributed in the northwestern region of china. The main medicinal active ingredients are matrine and sophocarpine. Modern pharmacological research shows that the above two alkaloids have anti-infective, anti-cancer function, and can enhance immunity. So they are widely used to treat women reproductive tract infection, viral hepatitis, tumor, cancer, asthma and other diseases. However, the extraction industry of Sophora alopecuroides L. is still in the early stage of development, and many of processes are less practical. Therefore, the extraction processes of matrine, sophocarpine are discussed, and the dynamic models under different conditions are established by studying on seeds of Sophora alopecuroides L.. As a result, the data is acquired to provide a reliable basis for the industrialization amplification.The extraction process and the optimum conditions are determined by using single factor experiments and orthogonal experiments. When the best acids extraction process is the extraction temperature 338K, diameter 0.15mm, extraction time 13 hours, sulfate solution pH 0.8, solvent-fold 20:1, the content of overall alkali (matrine) is 24.39% and yield is 69%; When the optimistic alkyd ultrasonic extraction process is the extraction temperature 298.5K, particle size 0.25mm, extraction time 55min, acid concentration 70%, ultrasound frequency 500KHz, field 120W/m2, the content of overall alkali (sophocarpine) is 34.42%, and yield is 65%.Hollow-fiber ultrafiltration membrane is used to purify acids extraction solution. Meanwhile, the separating effects of different polysulfone membrane on proteins, tannins and polysaccharides are compared, and the results show that the best parameters are operating pressure 0.3MPa, temperature 45℃, ultrafiltration concentration 6g/L.N0.001x8 cation-exchange resin is screened from varieties of resins, which has both big adsorption capacity and high desorption rate. Furthermore, the process and the parameters are optimized.The purification process of matrine and sophocarpine with Column chromatogramphy is optimized. The best purification conditions of matrine are elution rate 1.5mL/min, feeding 0.3g, column high 300mm, crystallization temperature 10℃; The best purification conditions of sophocarpine are the elution rate 2.5mL/min, sample volume 5mL, sample concentration 62% and adding water volume 0.8mL. By HPLC tests, the crystal purity of matrine and sophocarpine are respectively 99%, 98%.The different extraction dynamic models are established under different conditions, and the transfer mass mechanism is theoretically discussed. The results show that equilibrium constant (1.888×10-3) of ultrasonic extraction is 27.4 times higher than one (6.89×10-5) of conventional extraction, and the transfer mass coefficient of ultrasonic extraction is nearly two orders of magnitude more than conventional extraction. |
