Although the traditional Chinese medicine (TCM) industry developed rapidly, most wastewater from its production was discarded without treatment. It is very significant to research and develop high-powered, simple, economical, efficient and stable TCM wastewater treatment technologies. The TCM wastewater is one kind of high strength organic wastewater with multi contamination and complicated component. At present, most TCM wastewater biological treatment processes are focused on easily biodegraded wastewater whose Chemical Oxygen Demand (COD) is less than 5000 mg/L. But the COD of the wastewater discharged from Harbin Second TCM Factory was more than 18000 mg/L. A model project with two-phase anaerobic digestion process was developed to treat the high concentration and refractory organic wastewater discharged from Harbin Second TCM Factory. The results showed that the COD average removal of total process was around 94.06%. Therefore, the two-phase anaerobic digestion process can conquer this problem. This paper participated in the design research of the two-phase anaerobic digestion process project treating high concentration and refractory wastewater discharged from Harbin secondary TCM Factory, did the research for the microorganism of the acidogenic phase and the ethanogenic phase, and enriched and separated the germs. Depend on the shape of microbial cells and its characteristics, we can know how the microbe works and how they influence the reactor systems in the acidogenic phase and the ethanogenic phase. This paper provided a reference for increasing efficiency and stability of the acidogenic phase and the ethanogenic phase. We separated 24 germs from the acidogenic phase reactor and 19 germs from the ethanogenic phase reactor in Harbin Second TCM Factory。According to the characteristics of the 24 germs from the acidogenic phase and that of the 19 germs from the ethanogenic phase, we chose the NO.24 germ from the acidogenic phase and the NO.11 ferm from the ethanogenic phase. And we did the research for the characteristics of the NO.24 germ and the NO.11 germ. The results showed that the NO.24 germ Gram-positive, non-motile, the cell size is 0.64μm×0.7μm, the growth of the NO.24 germ sustaining cycle is 45 h. The NO.11 germ is Grampositive, non-motile, and the cell size of the NO.11 germ is 0.64μm×0.7μm. The optimal ranges of the NO.11 germ are 37℃, pH 7 and NaCl 3%, and it is judged Methanococcus mazei.
