Ganoderma lucidum is a famous medicinal and edible fungus with special nutrition and medicinal values. Utilizing its ability to enrich Zinc and transform inorganic-Zinc to organic-Zinc, which will combine both its value and that of organic-Zinc's together, the medicinal value of Ganoderma lucidum is highly improved. Therefore, the development of Zinc-enriched Ganoderma lucidum could extend application and have highly social and economic benefits. A strain of Ganoderma lucidum with high Zinc resistance and more suitable for liquid fermentation was chosen.The effect of Zinc on the growth and metabolism of the above chosen strain was studied. The cultivation conditions for liquid seed and fermentation medium components and fermentation conditions for Ganoderma japonicum to enrich Zinc were also optimized. The results of this study were as follows:①Resistance to Zinc and the Zinc-enriched quantity of Ganoderma sp.,Ganoderma lucidum (Fr.)Krast and Ganoderma japonicum were different. Overall, Ganoderma japonicum was more suitable for organic carrier of Zn-enrichment. 500mg/L Zinc was the appropriate concentration for Ganoderma japonicum.②Lower extracellular polysaccharide and intracellular polysaccharide of Ganoderma japonicum in liquid medium with 500mg/L of inorgnic-Zinc concentration was observed while higher extracellular and intracellular polysaccharide in mycelium was obtained.③The cultivation conditions for liquid seed of Ganoderma japonicum was established. The higher biomass and density and smaller pellet size with liquid fermentation were obtained when the cultivation conditions were inoculum amount 12 pieces, initial pH7.10, rotation speed 179r/min, temperature 26℃and medium volume 80ml in 250ml shake flask.④The fermentation medium for Ganoderma japonicum to enrich Zinc was confirmed. The medium components were corn flour 38.0 g/L, saccharose 20 g/L, bean cake 8.5 g/L, Fe2(SO4)3 1.0 g/L, NH4Cl 2.0 g/L, CaCl21.0 g/L, MgSO41.0 g/L, KH2PO4 1.2 g/L. Using this fermentation medium, the ratio of Zinc-enrichment could reach 27.32%. And the ratio of Zinc-enrichment increased 6.32% compared with the original medium. ⑤The fermentation conditions for Ganoderma japonicum to enrich Zinc were established. The fermentation conditions were initial pH8.30, inoculum amount 11.1%, rotation speed 179r/min, temperature 26℃and medium volume 80ml/250ml. Using this fermentation conditions, the ratio of Zinc-enrichment could reach 30.85%.
