This paper inwestigate the determination of apple polyphenols in apple pomace by a new Folin Method and exploring its new biological function.Apple polyphenols was tested with standard sample,gallic acid.Static adsorption and desorption tests were employed to screen the best macroporous resin for the apple polyphenols;Based on physic and chemical properties and adsorption mechanism of macroporous resin, the single factor tests,response surface method was carded out to optimize the manipulation parameters of macroporous resin for the adsorption and desorption of apple polyphenols,its active components were identified by using HPLC,the conclusions were list as follows:1.Through static adsorption and desorption test,NKA-9 macroporous adsorption resin was the best macroporous resin for the apple polyphenols.2.Studied the NKA-9 resins adsorption characteristic,drawed up isothermal adsorpting curve at 25℃,and fited with the model.The exporimental datas of adsorption satisfied with the Langmuir isotherm model and coefficient R2=0.9902;The datas also fitted Freundlich isotherm model and decision coefficient R2=0.9563.The forecast static biggest quantity of adsorption was Q0=63.69 mg/g.3.Studied the NKA-9 resins adsorption charactedstic,drawed up adsorption kinetics curves at 25℃,and fited with the model.The exporimental datas of adsorption satisfied with the Langmuir model and coefficient R2=0.9299,calculated adsorption equilibrium constant of AP on NKA-9 resin.It explained that adsorption of NKA-9 resins belonged to slow adsorption.4.Feeding rate,concentration of feed and pH were three key factors,response surface method was applied to optimize the manipulation parameters of macroporous resin for the adsorption of apple polyphenols and the equation which showed the relationship between the dynamic adsorption quantity and the 3 factors is: Y=19.19+0.74A+2.33B-1.70A2+0.74B2;The optimum adsorption conditions was at the feeding rate of 1.14ml/min,concentration of 2.5mg/ml,pH 4.47.The maximum dynamic adsorption quantitywas 22.31mg/g in theory.5.Elution rate,elution concentration and eluant volume were key factors,response surface method experimental designs were applied to optimize the manipulation parameters of macroporous resin for the deadsorption of apple polyphenols and the equation which showed a dynamic deadsorption relationship with the 3 factors, Y=86.15-7.85A+0.65B+0.58C-6.03A2-1.73B2-1.45C2-0.41AB-0.51AC+1.36BC;The optimum deadsorption conditions was at 0.79ml/min of the elution rate,which elution concentration was 60.18%,eluant volume116.27ml.The maximum dynamic deadsorption rate was 88.2057%in theory.6.The puried AP determined by HPLC,gallic acid,phloridzin and ferulic acid were identified and quantificated in apple pomace.In addition,some experiments were carded out to illustrate its antioxidant, inhibitory activities in vitro.Polyphenols from apple pomace exhibited strong scavenging capacities towards superoxide anion radicals,generated by a pyrogallol autoxidation system,hydroxyl radicals generated by Fenton reaction and nitrite.The maximum scavenging percentage for the free radicals and nitrite inhibition indicated in the above reactive systems reached 97.56%,95.65%and 76.98%,respectively.Four antioxidants tested were conducted with peanut oil to inwestigate the antioxidant effect with the instrument of Rancima.The induction time of peanut oil samle with AP was24.95h,which was three times that of control oil samle.Compared to the oil samle with BHT and Vc,induction time was 78.12%and 23.12%longer respectively,which was longer than TBHQ.Results showed polyphenols with a strong antioxidant activity.Using filters the slip of paper take the bacteriostasis circle diameter as the level, inspected the different density apple polyphenol solution to food common pollution fungus-Escherichia coli inhibitory action.In 0.1%~0.5%density scope,the apple polyphenol solution concentration displays the very strong inhibitory action, moreover the inhibitory action and the solution concentration presented the correlational dependence.In order to explore the new bio-function of apple polyphenols,animal experiment was carded out with Kunming mice,which were the lead poisoned by intraperitoneal injection of the lead acetate(PbAc).The observation apple polyphenol to the lead contamination mouse in vivo lead content influence,after discovered the lead contamination,after the contamination mouse oral administration different density apple polyphenol,the blood lead,the liver lead content and the thighbone store up the level remarkably to reduce,but urinates the lead,the excrement lead level remarkable increasing,which illustrated that the apple polyphenols discharge lead oppressed.
