Study On The Compound Active Polysaccharides By The Ganoderma Lucidum Fermenting The Lycium Barbarum.

The active combinations of polysaccharides from Lycium barbarum fermented by Ganoderma luicidum were studied in this paper. Based on evaluating the scavenging ability of the hydroxyl free radicals, the ganoderma luicidum polysaccharide (GLP) and the lycium barbarum polysaccharide (LBP) were mixed in vitro to testify the feasibility and validity of compound polysaccharides. The results showed that the scavenging action of mixed polysaccharides in vitro was enhanced more than 10% related to the single polysaccharide.For further optimization study of fermentation media and conditions, the optimal results were as follow: 5%Lycium Barbarum, 0.225% MgSO4 , 0.159%KNO3 , 1% bran juice , 150r/min , 28℃, with inoculum size 10%(v/v) in 100mL/250mL conical flask and 7-day cultivation. Finally, the polysaccharides concentration achieved 2.831g/L in the fermentation juice, and the scavenging action of fermentation polysaccharides reached 41.30%.The polysaccharides were separated by the DEAE-cellulose. The results showed that the GLP had GLPⅠ,GLPⅡand GLPⅢ; the LBP had LBPⅠ,LBPⅡand LBPⅢ,LBPⅣ; the mixed polysaccharide(C) had CⅠ,CⅡ,CⅢ,CⅣ,CⅤand CⅥ; the fermentation polysaccharide(F) had FⅠ,FⅡ,FⅢ,FⅣand FⅤ. By the HPLC analysis, it showed evidently differences between the mixed in vitro polysaccharides and the fermentation polysaccharides on the composition, molecule mass, as well as the molecular structure.Based on the above studies, the development of fermentation juice product was carried out. The optimal fermenting conditions were as follows: 150 rpm , 28℃, 4-day cultivation ; the scavenging action of the hydroxyl free radicals by fermentation juice reached 56.65% . Finally, the good-taste, clear, transparent active compound health protection beverage was obtained.An elementary study has been made on the active combinations of polysaccharides by microorganism fermenting Chinese traditional medicines in this paper. It has provided research foundation on active substances of Chinese traditional medicines .
