Objective: To formulate quality standard and evaluation and have a preliminary assessment on decreasing the blood lipid of grape essence soft-gelatin capsule. The producing method includes steps mentioned below. The material was residue of grapes in winery; two compositions are grape peel and grape seed which were separated from waste residues of grape. The Oleanolic acid (OA) from crude extraction of the grape peel, the Oligomeric Proantho Cyanidins (OPC) from grape seed and the extraction also comprised grape seed oil as solvent, Together with some supplemental material, the grape essence soft-gelatin capsules were made ultimately. Methods:â‘ To obtain the extractum from grape peel by the method of alcohol extraction and purify the extractum.â‘¡The main component of grape essence soft-gelatin capsules was extracted from grape peel and seed, the solvent was grape seed oil and then the making prescription and the best method of making capsules were selected.â‘¢The content of OA and OPC were measured by HPLC and UV. Then the stability of products was observed primarily by accelerating-test, and formulated the quality standard.â‘£To establish the high cholesterol animal model and then divided into five groups randomly by the level of total cholesterol (TC), which group had 12 mice. They were hyperlipemia control, solvent control and high dosage, middle dosage and low dosage group. Intragastric administration to high dosage with the water,2% Tween-80 water to the solution control. High dosage, middle dosage and low dosage group were lavaged with capsule content solution of high, middle and low concentration respectively. Every group's volume was 1ml/100g for 25 days, one time per day. Five groups fed with high fat food and weighed regularly. To abrosia 12 hours before finishing the experiment and got the blood from medial angle of eye. Then centrifuging and separating the blood serum to detect the content of total cholesterol (TC), triglyeride(TG) and high density lipoprotein(HDL).Finally investigated the decreasing blood lipid effects of the grape essence soft-gelatin capsule. Results:â‘ The grape essence soft-gelatin capsule could be produced successfully from the extraction of grape peel and seed. The best prescription was grape seed oil 72.5%, OA powdered extract 17.5%, OPC powdered extract 10.0%, beeswax 3%,TBHQ 0.01%.â‘¡The method of measuring the content of OA with HPLC and OPC with UV was feasible and accurate, and could establish the quality criteria. The result is OA 3.68% and OPC 9.49%.â‘¢According to the animal experiment, both middle and high level dose group could decrease high cholesterol in animal model's serum, and the difference had statistical significance. Conclusion: The study demonstrates that the prescription of grape essence soft-gelatin capsule is reasonable, and the content is steady; according to the function appreciation program and analysis process of health food, the animal experiment's result shows that: compared with the hyperlipemia control and solvent control group high and middle concentration group of TC have statistical significance. As can we see that: the function of decreasing the serum cholesterol of grape essence soft-gelatin capsule, it has certain prospect of market development.
